隐藏嗜酸菌Acidiphilium cryptum JF-5分泌蛋白质组的分析

         

摘要

[目的]对隐藏嗜酸菌(Acdiphiliumc cryptum JF-5)全基因组的3 063个氨基酸序列进行预测,分析其分泌蛋白质组的构成.[方法]利用信号肽预测软件SignalP v3.0、跨膜螺旋结构预测软件TMHMM v2.0 、RR-motif信号肽预测软件TatP v1.0、脂蛋白质信号肽预测软件LipoP v1.0、非经典分泌蛋白质预测软件SecretomeP和亚细胞定位软件Psortb v2.0.4对Acidiphilium cryptum JF-5全基因组的3 063个氨基酸序列进行预测分析.[结果]在隐藏嗜酸菌Acidiphilium cryptum JF-5中有444个蛋白质为分泌蛋白质,其中分泌型信号肽148个,RR-motif亚组型信号肽27个,脂蛋白质信号肽22个,无Prepiplin-like信号肽,非经典分泌蛋白质247个;另外,对分泌型信号肽的长度分布、氨基酸使用频率和酶切位点的氨基酸使用频率作出了统计分析.[结论]研究结果为进一步研究嗜酸菌Acidiphiliumc cryptumJF-5的基因功能提供了丰富的信息基础.%[Objective ] The component of the secreted protein from Acidiphilium cryptum JF-5 genome was analyzed through the prediction of 3063 amino acid sequences in it. [Method] The internet-based software SignalP v3.0, TMHMM v2.0, TatP vl.0, LipoP vl.0.SecretomeP and Psortb v2.0.4 were combined to predict the signal peptides from the 3063 ORFs in Acuiiphiiium cryptum JF-5. [ Results] The results showed that total 444 proteins tested in Acidiphilium cryptum JF-5 were the secreted protein, among which, 148 proteins was Sec-type signal peptide; 27 proteins, RR-motif signal peptides; 22 proteins, lipoprotein signal peptides, 247 proteins, non-classical protein and no Prepilin like signal peptide was found. Additionally, the length of the Sec-type signal peptide, the frequency of amino acid working, the working fre quency of particular amino acid in restriction sites were statistically analyzed. [ Conclusion] The rich information for the study on the function of Acidiphilium cryptum JF-5 genome was provided.
