药用植物青天葵叶片原生质体的制备与纯化

         

摘要

[Objective] To study the preparation and purification method of protoplast from Nervilia fordii (Hance) Schltr. Leaves. [Method] Taking the yield and survival rate of protoplast as index, the effects of different factors such as pretreatment, enzymolysis method, enzymolysis temperature, rotate speed of centrifugation, illumination, and sieve mesh number on the separation and purification of protoplast from N. Fordii leaves were studied. [Result] The optimum process for protoplast isolation was obtained as follows; N.fordii leaves were first pretreated for 1 h in CPW solution containing 13% mannitol,and then incubated at (25 ± 1)℃ in enzyme solution under dark conditions; after the enzymolysis,the enzyme liquid was filtrated through 200 mesh screen and centrifuged for 8 min at the speed of 800 r/min. Repeat the filtration and centrifugation once before microscopic examination. Under the above conditions, a large amount of complete and clear protoplast with few cell debris was obtained. [ Conclusion ] The preparation system has been successfully established and laid a good foundation for establishing protoplast regeneration system from N. Fordii leaves.%[目的]研究青天葵原生质体的制备与纯化方法.[方法]以青天葵新鲜叶片为试验材料,以青天葵叶片原生质体产量及原生质体存活率为考察指标,研究了预处理、酶解方式、酶解温度、离心转速、光照、筛网目数等因素对青天葵原生质体分离纯化效果的影响.[结果]适宜青天葵叶片原生质体游离的条件是用浓度13%甘露醇溶液预处理叶片1h、酶解温度(25±1)℃、黑暗、静置;原生质体纯化采用离心沉淀法,以200目筛网过滤后离心8 min (800 r/min),操作2次,可得到大量完整清晰、细胞碎片少的原生质体.[结论]该方法获得了制备青天葵原生质体的理想条件与参数,为建立青天葵原生质体再生体系提供了依据.
