根霉Yc1发酵豆渣产纤溶酶的研究

         

摘要

[目的]研究根霉Yc1以豆渣为原料经液态发酵产生纤溶酶的发酵条件.[方法]采用单因素试验与正交设计方法对根霉Yc1培养基及发酵工艺条件进行优化.[结果]根霉Yc1发酵产酶的适宜培养基组成为:麸皮5.00%,豆渣粉3.00%,ZnCl_2 0.03%,MgSO_4 0.15%,Na_2HPO_4 0.05%;适宜的发酵工艺条件为:发酵温度32 ℃,初始pH值7.0,250 ml三角瓶装液量60 ml,摇瓶转速180 r/min.在优化条件下培养60h,纤溶酶的发酵效价可达176.2 U/ml,比优化前提高92.3 U/ml.[结论]以豆渣为主要原料所产的纤溶酶可能成为一种安全、低毒的溶栓剂,其生产成本低,可为大豆综合利用提供新的思路.%[Objective]The optimization of liquid state fermentation conditions of fibrinolytic enzyme produced by Rhizopus sp. Yc1 from soy residue were studied. [Method]The optimum fermentation conditions were determined by monofactor test and orthogonal design . [Result] The optimum medium consisted of wheat bran 5.0%,soy residue flour 3.0%,ZnCl_2 0.03%,MgSO_4 0.15%,Na_2HPO_4 0.05%;and the medium initial pH 7.0, 60 ml medium in 250 ml Erlenmeyer,rotation speed 180r/min for 60h at 32 ℃.The fibrinolytic enzyme productivity reached 176.2 U/ml on optimized condition,which is more than 92.3 U/ml that of the original conditions.[Conclusion]The fibrinolytic enzyme produced from soy residue would be a safety cellosolve,which with high activity, low side reaction and low cost.The study provides soy residue as produce material with a new direction.
