The toxicity effects of glufosinate ammonium to Tetrahymena thermophila were investigated from the viewpoints of cell prolif-eration and DNA damage .The growth curves of 96 hours were plotted by counting the cells every 12 hours;the tail length , tail moment and Olive tail moment of Tetrahymena thermophila were measured by Single Cell Gel Electrophoresis (SCGE).The results of growth curve test showed that there was a significant difference between drug groups and control group in cell density ( P<0.01 ) .According to SCGE data , the tail length , tail moment and Olive tail moment of the tested series were all significantly larger and higher than those in the control group ( P<0.01 ) , the DNA damage level was higher when toxic compound become heavier ( P<0.01 ) .The experi-mental concentration of glufosinate ammonium caused damages of cell DNA in varied extents , which reflects the acute and genetic toxic-ity of it to Tetrahymena thermophila .%从细胞增殖和DNA损伤角度研究不同浓度的草铵膦对嗜热四膜虫( Tetrahymena thermophila)的毒性作用。采用12 h间隔细胞计数绘制96 h生长曲线,单细胞凝胶电泳技术检测受试四膜虫的尾长( TL)、尾矩( TM)及Olive尾矩( OTM)。生长曲线绘制实验表明:草铵膦处理组与空白组种群细胞数目差异显著( P<0.01)。单细胞凝胶电泳实验表明:染毒各组的细胞TL、TM、OTM与对照组相比,均有显著增大( P<0.01) ,DNA损伤级别随着染毒浓度增大而升高( P<0.01)。实验浓度的草铵膦对细胞DNA均造成了不同程度的损伤,反映了其对四膜虫具有急性遗传毒性。
