番茄红素对急性肺损伤大鼠肺毛细血管膜通透性及骨形态生发蛋白-4基因表达的影响

         

摘要

Objective:To observe the effects of lycopene (Lye)on pulmonary capillary permeability and gene expression of bone morphogenetic protein-4(BMP-4)mRNA after acute lung injury(ALI)in rats and investigate the protection of lycopene on ALI.Methods: Sixty rats were randomly divided into 3 groups: control group ( Ⅰ), ALI group ( Ⅱ )and ALI+Lyc group( Ⅲ ).Animal models of ALI induced by HCL inhalation in rats were made to determine the capillary permeability, lung wet/dry weight ratio, myeloperoxidase (MPO)acticity of lung tissue after 6h and the changes of the expression of BMP-4 mRNA in lung tissue.Results: Compared with group Ⅰ , in group Ⅱ,extraction of Evans blue in lung tissue increased very obviously (P<0.01) ,lung wet/dry weight ratio, MPO of lung tissue increased obviously (P<0.05), the expression of BMP-4 mRNA in lung tissue up-regulated obviously (P<0.05).Compared with group Ⅱ,in group Ⅲ .extraction of Evans blue,lung wet/dry weight ratio,MPO of lung tissue decreased obviously (P<0.05),the expression of BMP-4 mRNA in lung tissue down-regulated obviously(P<0.05).ConcIusion:Lycopene show a marked reductive effect on the capillary permeability, lung wet/dry weight ratio,MPO activity of lung tissue and suppress the up-regulation of the expression of BMP-4 mRNA,relieve capillary membrane injuries during prophase and may improve the restoration of lung tissue during anaphase of ALI in rats,has protective effects on lung in ALI rats.%目的:观察番茄红素(Lyc)对急性肺损伤(ALI)大鼠肺组织毛细血管膜通透性及骨形态生发蛋白-4(BMP-4)mRNA基因表达的影响,探讨其对ALI的保护作用.方法:将Wistar大鼠随机分成3组:对照组(Ⅰ组)、ALI组(Ⅱ组),ALI+Lyc组(Ⅲ组),每组20只,每组中10只处死前注射伊文思蓝.从模型制备前1周开始,Ⅰ组和Ⅱ组用橄榄油、Ⅲ组用橄榄油稀释的番茄红素树脂(1g/L)以5ml/kg的剂量灌胃,1次/d.制备酸吸人大鼠ALI模型,测定ALI后6h肺毛细血管通透性、肺湿干质量比、肺组织髓过氧化物酶(MPO)活性及BMP-4 mRNA的表达.结果:与Ⅰ组比较,Ⅱ组肺组织中伊文思蓝提取量显著增加(P<0.01),肺湿干质量比值、MPO活性显著升高(P<0.05),BMP-4 mRNA表达水平显著上调(P<0.05);与Ⅱ组比较,Ⅲ组肺组织中伊文思蓝提取量显著降低(P<0.05),肺湿干质量比值、MPO活性明显均不同程度降低(P<0.05),BMP-4 mRNA的表达水平显著下降(P<0.05).结论:番茄红素降低酸吸入致ALI大鼠的肺毛细血管通透性、肺湿干质量比及肺MPO活性,抑制BMP-4 mRNA表达,减轻ALI大鼠早期肺泡毛细血管膜损伤并可能改善后期肺组织的修复,具有肺保护作用.
