目的 研究生长追赶宫内发育迟缓(CG-IUGR)大鼠脂肪组织中巨噬细胞浸润及分泌炎症细胞因子水平改变,以探讨CG-IUGR大鼠脂肪组织炎症反应与胰岛素抵抗的关系.方法 SD大鼠怀孕期间全程给予正常孕鼠食量的30%以限制饮食,并通过增加仔鼠哺乳期营养摄入建立CG-IUGR大鼠模型.以8周龄雄性CG-IUGR仔鼠为研究对象,取附睾脂肪组织做石蜡切片,通过免疫组化染色鉴定脂肪组织中浸润的巨噬细胞.大鼠原代成熟脂肪细胞和巨噬细胞用Transwell系统行共培养,ELISA法测定细胞培养液上清中炎症因子IL-6和TNF-α表达水平.结果 8周龄CG-IUGR仔鼠体质量、体质指数均高于对照组(P均<0.01);脂肪组织中巨噬细胞浸润较对照组大鼠增多;细胞共培养体系培养液中IL-6、TNF-α水平较对照组增高(P均<0.01).结论 CG-IUGR大鼠脂肪组织存在巨噬细胞浸润和慢性炎症反应,参与机体胰岛素抵抗的形成.%Objective To investigate the changes of macrophages infiltration in the adipose tissue of intrauterine growth retarded rats with catch-up growth (CG -IUGR), and to investigate the secretion of proinflammatory cytokines in order to explore the impact of inflammation in adipose tissue of CG-IUGR rats with insulin resistance. Methods Rats in food-restrict group were fed 30% of intake of food comsumed by control group during pregnant, and the newborns were fed more breast milk to ensure their catch-up growth. Adipose tissue was isolated from epididymal fat pad of 8 week-old rats and paraffin section was stained by immu-nohistochemistry. Primary cultured rat peritoneal macrophages were co-cultured with primary cultured rat adipocytes for 24 hours intranswell inserts. The levels of IL-6 and TNF-ainthe supernatant of co-cultured cells were detected by ELISA. Results The weight and body mass index (BMI) of 8 week-old CG-IUGR rats were higher than those of controls (P<0.01). The accumulation of macrophages in adipose tissue of 8 week-old IUGR rats was more than that in control rats. The levels of proinflammatory cytokines IL-6 and TNF-a in the supernatant of co-cultured cells of CG-IUGR rats were higher when compared with the controls (both P<0.01). Conclusions Adipose tissue of CG-IUGR rats is characterized by macrophage infiltration and these macrophages are an important source of inflammation in adipose tissue.
