珍珠层组织工程支架材料的研制及表征

         

摘要

Objective To develop a nacre-derived scaffold material for hone tissue engineering and test its properties. Methods The nacre powder and Poly-DL-lactic Acid (PDLLA) were proportionally mixed to prepare the nacre/poly-DL-lactic acid artificial hone (NPAB) hy phase separation/salt leaching technique. The characteristics of the NPAB, such as surface morphologies, hydrophilicity, mechanical properties, and in vitro degradation parameters were determined. Results The average porosity rate of the NPAB was more than 85%. The big pore size between ranged between 150 and 300 μm and the small pore size ranged from 10 to 30 μm with effective connectivity. The hydrophilicity, mechanical properties, degradability of the scaffolds with different nacre proportion were much better than those of the pure PDLLA. The compressive strength of Nacre/PDLLA(3/2) group reached to (3.871 7 + 0. 198 0) MPa, which was similar to cancellous bone. Statistical differences (P<0. 05) were found in mass loss and solution pH between OPAB and pure PDLLA groups. Conclusion The properties of Nacre/PDLLA made by Phase separation/ salt leaching technique were superior to those of pure PDLLA. Among them, the mechanical properties of Nacre/PDLLA scaffold with a proportion of 3:2 performed best, which met the requirements of bone tissue engineering scaffolds.%目的 自主研发贝壳珍珠层骨组织工程支架材料并检测其各项基本性能.方法 采用相分离/盐析复合工艺将珍珠层粉、消旋聚乳酸(PDLLA)不同比例复合,制备珍珠层聚乳酸人工骨(NPAB),检测其微观形貌、孔隙率、亲水性、生物力学强度、体外降解性能等表征材料的各项属性.结果 制备的NPAB大孔孔径150~250 μm,小孔孔径10~30 μm,孔间连通性好,孔隙率≥85%,亲水性、力学强度、体外降解性均优于对照组纯聚乳酸组.珍珠层/PDLLA比例3∶2组的压缩强度达(3.871 7±0.198 0)MPa,与松质骨相近,高于其余各组(P<0.05).NPAB质量损失率和pH值与纯聚乳酸组间存在统计学意义(P<0.05).结论 相分离/盐析法制得的不同比例珍珠层/PDLLA材料的各项性能均优于纯PDLLA组,其中珍珠层/PDLLA以3∶2复合组的性能最优,可作为良好的骨组织工程支架材料.
