Objective To study the expression and clinical significance of the chemokine receptor CXCR3 on T lymphocytes from peripheral blood cells in patients with acute leukemia. Methods Flow cytometry and direct immuofluorescence staining were used to detect the expressions of CXCR3 on T lymphocytes from peripheral blood cells in 133 cases of AL and 33 cases of normal controls. Results The positive expression rate of chemokine receptor CXCR3 on the surface of T lymphocyte from the peripheral blood cell in the patients with AL was (28. 24 ± 18. 14)%, significantly higher than the control groups (12. 19 ± 3. 89)%(P<0. 05). Among different subtypes of AL, the expression rate in B-ALL was the highest (40. 14 ± 17. 56)%, while T-ALL, with expression rate of (14. 06 ± 8. 29) %, was only slightly higher than the normal control group. The CXCR3 expression on T lymphocytes in AL patients with ex-tramedullary filtration was(34. 54 ± 19. 87) %, and was significantly higher than those without extramed-ullary filtration (20. 44 ± 12. 15) %(P<0. 01). However, the expression of CXCR3 on the T lymphocytes from the peripheral blood cells in AL patients was not correlated to the WBC count, the percentage of blast cells, as well as CD3 positive rate. Conclusion The CXCR3 expression on T lymphocytes from the peripheral blood cells in patients with AL might be closely associated with migration of the leukemic cells and extramedullary organ infiltration.%目的 探讨急性白血病(AL)患者外周血T淋巴细胞趋化因子受体CXCR3表达及其临床意义.方法 采用流式细胞术结合直接免疫荧光法检测133例AL以及33例对照组外周血T淋巴细胞CXCR3表达.结果 AL患者外周血T淋巴细胞表面趋化因子受体CXCR3的表达阳性率明显高于对照组,分别为(28.24±18.14)%和(12.19±3.89)%(P<0.05).在各型AL中,以B-ALL的表达率最高(40.14±17.56)%,而T-ALL型为(14.06±8.29)%,仅略高于正常对照(P>0.05);AL髓外浸润患者外周血T淋巴细胞CXCR3的表达为(34.54±19.87)%,明显高于非髓外浸润患者(20.44±12.15)%(P<0.01).但AL患者外周血T淋巴细胞CXCR3的表达与白细胞总数、幼稚细胞百分率及CD3阳性率表达无关.结论 AL外周血T淋巴细胞CXCR3的表达可能与白血病的迁移、浸润密切相关.
