TLR9在结直肠癌中表达特点的研究

         

摘要

Objective:The role of toll-like receptor 9 (TLR9) in colorectal cancer (CRC) is still unknown.This study investigated the expression of TLR9 in colon tissues from mice under the process of CRC formation as well as from clinical CRC samples.Methods:The model of colitis-associated CRC was used, which involves treating C57BL/6 mice with azoxymethane (AOM) injection followed by dextran sulfate sodium (DSS).Mouse colon tissues were collected on the early time of tumorigenesis (day 14) and TLR9 gene expression was determined by qRT-PCR.mRNA and protein expression of TLR9 in 26 fresh human CRC samples were determined by qRT-PCR and Western-blot, respectively.Results: When compared with controls, the level of TLR9 mRNA was significantly decreased in mouse colon tissues on the early time of the formation of CRC (P<0.05).Both the protein and mRNA levels of TLR9 were significantly reduced in human fresh CRC tissues (P<0.05).Conclusion:The decreased expression of TLR9 in colorectal tissues is involved in process of CRC formation, and TLR9 may have suppressive effect and therapeutic value for CRC.%目的:探讨TLR9 (Toll-like receptor 9)在小鼠结直肠癌(Colorectal cancer,CRC)形成早期过程中及在临床CRC样品中的表达水平.方法:通过予以C57BL/6小鼠腹腔注射氧化偶氮甲烷(Azoxymethane, AOM)后接以3轮的葡聚糖硫酸钠(Dextran sulfate sodium, DSS)处理建立炎症相关性结直肠癌模型,采集肿瘤形成早期 (第14天) 结直肠组织,通过qRT-PCR方法检测TLR9基因在小鼠结肠中的表达.另外,通过Western-blot和qRT-PCR检测26对临床新鲜结直肠癌组织及相应癌旁组织TLR9的表达水平.结果:与对照组相比,TLR9在小鼠结直肠癌形成早期及人结直肠癌组织中的表达均呈显著性下降(P<0.05).结论:结直肠中TLR9低表达可能是CRC的促因之一,TLR9可能具有抑制CRC的发生发展和治疗CRC的潜能.
