索氏提取法提取裂殖壶菌粉油脂参数的优化

         

摘要

Schizochytrium has become one of the research hotspots of marine fungi for its high contents of DHA , while the oil grease extraction efficiency of schizochytrium powder is not high .The soxhlet extraction method is recognized as an effective way to improve the extraction efficiency .Under the condition of two mixed solvent ,the article discusses the effect of extraction time ,amount of powder and temperature on grease extraction from the new and old schizochytrium powder . In addition , the optimal extraction condition has been determined through the experiment and the orthogonal experiment .The results show that when the organic solvent is mixed by equal volume of chloroform and methanol ,the amount of algae powder is 1 .0 g ,extraction time is 2 .5 h ,and temperature is 95℃ ,the extraction rate of grease is the highest ,being 81 .70% for new and 47 .43% for old powder material . Through repeated experimental verification under the optimal condition , the article finds out that this optimized processing is effective in improving grease extraction rate and powder discrimination , which will provide useful references for grease extraction and future industrial application .%裂殖壶菌作为富含DHA的海洋真菌成为研究热点,但其干粉的油脂提取效率并不高,索氏提取法是能够提高油脂得率的方法之一。通过实验探讨了两种不同混合溶剂条件下,提取时间、藻粉量、水浴温度等因素对新旧两份裂殖壶菌藻粉材料油脂提取的影响,并通过正交试验分别确定了其最佳提取条件组合:有机萃取溶剂为1∶1等体积氯仿—甲醇混合溶剂,藻粉量1.0g,萃取时间2.5h,水浴温度95℃,得到新、旧藻粉的最高粗油脂提取率分别为81.70%和47.43%。在最佳条件下进行重复实验验证,发现优化后的索提方法油脂提取率高,可以有效甄别新旧菌粉,将为裂殖壶菌油脂提取工艺的优化及实际工业应用提供实验依据。
