Objective To investigate effects of polysaccharides on the solubility and pharmacokinetics of baicalin. Methods Baicalin in samples in vitro and in vivo were determined by high performance liquid chromatography ( HPLC ) . By comparing the effects of different polysaccharides on baicalin, the most effective polysaccharides were selected and administered to SD rats for pharmacokinetic experiments. The pharmacokinetic parameters were calculated by DAS 2. 0. Results Ganoderma lucidum polysaccharides affected the solubility of baicalin significantly, and the solubility of baicalin in Ganoderma lucidum polysaccharides mixture increased from 6. 68 μg/mL to 92. 93 μg/mL. Compared with the group using baicalin alone, AUC0-t , Cmax , Tmax of baicalin in combined group increased by 27. 16%, 29. 67%, and 52.03%, respectively. CL decreased by 19. 47%. Conclusion Ganoderma lucidum polysaccharide could increase the water solubility of baicalin, and was helpful to promote the absorption of baicalin.%目的 考察中药多糖对黄芩苷溶解度及其体内药动学的影响.方法 采用高效液相色谱法测定体内及体外样品中黄芩苷的质量分数;通过比较不同多糖对黄芩苷溶解度的影响,选取影响最大的多糖进行SD大鼠药动学试验,DAS 2.0软件计算得到药动学参数.结果 灵芝多糖对黄芩苷溶解度的影响最明显,黄芩苷与灵芝多糖混合前后在水中的溶解度由6.68μg/mL增至92.93μg/mL.与单用黄芩苷组比较,灵芝多糖组的AUC0-t增加了27.16%,Cmax增加了29.67%,达峰时间Tmax延长了52.03%,而CL减少了19.47%.结论 灵芝多糖可增加黄芩苷的水溶性,促进黄芩苷的吸收,提高其生物利用度.
