大麦黄矮病毒株系间分子变异与系统进化关系研究

         

摘要

Forty-five complete sequences of barley yellow dwarf virus ( BYDV) genome were analyzed with multiple alignments, and the results showed that there are two different genome structure types of BYDV. Two phylogenetic trees of BYDV were established based on 45 complete genomes and 166 coat protein (cp) genes sequences, and the results showed that PAV strains could be divided into three subgroups, and the PAV isolates from China were different from the isolates from Europe or America. The MAV strain could also be divided into three subgroups, and the MAV ( one of three subgroups in USA) held high similarity with GAV (in China) in genome sequences. 33 CP amino acid sites were found completely conserved in the same strains but were highly variable among different strains. Strain-specific sites of PAV, SGV, GPV were found by sequencing analysis, but no strains-specific variable site was found between GAV and MAV, indicating that the GAV might be one subgroup of MAV.%对已公布的45条大麦黄矮病毒( Barley yellow dwarf virus,BYDV)全基因组进行比对分析,表明BYDV具有2种基因组结构类型.依据45条全基因组序列和59条外壳蛋白(CP)基因序列分别建立BYDV系统进化树,表明PAV株系可分为3个亚组,且发现中国的PAV与欧美的PAV间存在明显的分子差异.MAV株系也可分为3个亚组,其中1个MAV亚组与中国的GAV株系具有较高的相似性.找出了在株系内完全保守同时在株系间发生变异的CP氨基酸位点33个,明确了PAV,SGV,GPV等3个株系CP氨基酸序列中具有株系特异性的变异位点,发现MAV和GAV在这33个变异位点处全部相同,无特异性变异位点,推测GAV株系可能是MAV的1个亚类.
