柱前衍生-反相液相色谱法检测花生粕中的18种氨基酸含量

         

摘要

We described a method for content determination of 18 kinds of amino acids by reversed-phase high performance liquid chromatography in the paper. Taking phenylisothiocyanate (PITC) as the precolumn derivatizing agent, the method could well separate the 18 kinds of amino acids in 45 minutes by the steps of binary gradient elution using Venusil column at 40 'C and up detection at 254 run. The results showed that the amino acid derivatization solutions kept stable in 24 hours; the concentration of amino acids was in good linear relationship with the peak area; the linear correlation coefficients of the 17 kinds of amino acids except cysteine (R2=0.988 3) were lager than 0.990 0; RSDs were 1%~3%; the recoveries of standard addition of the 18 kinds of amino acids were 96.5% ~ 102.9%; and the results had well repeatability and stability. The method was applied to determine the content of amino acids in peanut meal, and obtained satisfactory results.%探讨了一种利用反相高效液相色谱测定18种氨基酸的方法.以异硫氰酸苯酯为柱前衍生剂,用Venusil液相色谱柱在40℃下进行二元梯度洗脱,于紫外254 nm处检测,18种氨基酸在45 min内获得较好分离.结果显示:氨基酸衍生溶液在24h内保持稳定,氨基酸浓度与峰面积呈现较好的线性关系,除半胱氨酸(R2=0.988 3)外,其他17种氨基酸的线性相关系数均大于0.990 0,相对标准偏差为1%~3%,18种氨基酸的加标回收率在96.5%~102.9%,具有良好的重复性与稳定性.
