柱前衍生法测定氨基酸含量试验中AccQ.Flour衍生试剂用量研究

         

摘要

目的:探索不同浓度氨基酸标准溶液的最佳衍生试剂用量.方法:通过取20μl衍生试剂所能衍生的氨基酸标准液的最高浓度样品,依次减少衍生试剂用量,根据数理统计学分析,比较各组峰面积与标准20μl所得峰面积有无显著性差异,探索各浓度范围的氨基酸标准溶液所需的最佳衍生试剂用量.结果:在浓度为0.75μmol/ml的氨基酸标准溶液中,加入17μl衍生试剂所得峰面积与标准20μm相比,无显著性差异;在浓度为0.5μmol/ml的氨基酸标准溶液中,加入15μl衍生试剂所得峰面积与标准20μl相比,无显著性差异.结论:当氨基酸浓度≤0.5μmol/ml时,衍生试剂用量为15μl即可充分衍生;对于浓度≤0.75μmol/ml的氨基酸标准溶液,17μl衍生试剂为最佳用量.%Objective: To find out the best dosage of derivative reagent which used for the different concentrations of standard solutions of amino acids. Methods: Taked the highest concentration of amino acid sample which can derived by 20μl derivative reagent, and reducted derivative reagent dosage followed to find out the the best dosage of derivative reagent which used for the different concentrations of standard solutions of amino acids according to mathematical statistical analysis. Results: The peak area of the 0.75 μmol/ml amino acid standard solution derived by 17μl derivative reagent was not significant different with the peak area derived by 20 μl derivative reagent. The peak area of the 0.5 μmol/ml amino acid standard solution derived by 15 μl derivative reagent was not significant different with the peak area derived by 20 μl derivative reagent. Conclusion: The best dosage of derivative reagent which used for the standard solutions of amino acids which concentration not over 0.5 μmol/ml is 15μl; the best dosage of derivative reagent which used for the standard solutions of amino acids which concentration not over 0.75 μmol/ml is 17μl.
