中药脉平片质量控制标准的提高

         

摘要

Objective To establish a quality standard for Maipingpian,a Chinese medicine.Methods Thinlayer chromatography (TLC) was applied to the identification of Ginkgo biloba leaves (EGb),and TLC of emodin and physcion was improved.Total flavonol glycosides determined by high performance liquid chromatography (HPLC) was used with a photo-diode array detector.Results TLC of Ginkgolide A,EGb,emodin and physcion had distinct characteristic spots of separation and there was no interference in the negative comparison.The linear range of quercetin was between 0.029-0.588 μg (r=0.999 7);Kaempferol was between 0.062--0.411 μg (r=0.999 8);Isorhamnetin was between 0.057-0.566 μg (r=0.999 8).The mean recovery of quercetin was 97.22%,RSD=l.28%;Kaempferol was 98.15%,RSD=1.40%;Isorharrnetin was 97.16% and RSD=l.47%.Conclusion TLC and HPLC are accurate,reliable and simple in operation,which can be used for the quality control of Maipingpian.%目的 通过进一步完善脉平片成分的测定,建立更全面的质量控制标准.方法 采用薄层色谱法(TLC)对处方中银杏叶提取物进行定性鉴别,并对大黄素、大黄素甲醚的TLC法进行改进;采用高效液相色谱法(HPLC)对脉平片中总黄酮醇苷进行定量测定.结果 银杏内酯A、银杏叶提取物、大黄素、大黄素甲醚的薄层色谱特征斑点分离清晰,阴性对照无干扰;槲皮素在0.029-0.588 μg范围内所呈的线性关系良好,r=0.999 7;山柰素在0.062-0.411 μg范围内所呈的线性关系良好,r=0.999 8;异鼠李素在0.057-0.566 μg范围内所呈的线性关系良好,r=0.999 8.槲皮素平均加样回收率为97.22%,相对标准偏差(RSD) =1.28%;山柰素平均加样回收率为98.15%,RSD=1.40%;异鼠李素平均加样回收率为97.16%,RSD=1.47%.结论 TLC法和HPLC法操作简便、结果准确、重复性好,可用于控制脉平片的质量.
