人脐带间充质干细胞移植治疗大鼠压力性尿失禁

         

摘要

目的 探讨人脐带间充质干细胞(HUMSCs)移植治疗大鼠压力性尿失禁(SUI)的作用机制及有效性和可行性.方法 建立SUI大鼠模型,模型大鼠分为HUMSCs治疗组8只(A组,大鼠尿道周围注射HUMSCs约1×107个细胞)和实验对照组8只(B组,相同部位注射等量生理盐水),另取6只正常大鼠作为正常对照组(C组).建模后4周,将HUMSCs移植注射于大鼠尿道周围,在移植注射前1周及移植注射后6周分别测量所有大鼠最大膀胱容量(MBC)和漏尿点压力(LPP);行免疫组织化学检查,分析大鼠尿道括约肌中结蛋白、重链肌球蛋白(MHC)和蛋白基因产物9.5 (PGP9.5)的表达.结果 HUMSCs治疗组大鼠MBC、LPP测量值均较移植治疗前明显增加,差异有统计学意义(P<0.01);与实验对照组的MBC、LPP测量值相比显著增加,差异有统计学意义(P<0.01).免疫组织化学检查显示HUMSCs治疗组大鼠尿道括约肌中结蛋白、MHC、PGP9.5含量均较实验对照组明显增多,差异有统计学意义(P<0.01).结论 HUMSCs移植治疗SUI大鼠有效、可行,HUMSCs通过修复损伤的大鼠尿道括约肌和周围神经来提高控尿功能.%Objective To explore the mechanism,effectiveness and feasibility of human umbilical cord mesenchymal stem cell (HUMSCs) transplantating therapy in rats with stress urinary incontinence (SUI).Methods A SUI rats model were established and SUI rats were divided into two groups,8 rats in the HUM-SCs treatment group and 8 in the experimental control group,with other normal rats in a normal control group.In the HUMSCs treatment group,HUMSCs were injected into the rats' urethra at the 4th week,after the SUI rats model was established.The leak point pressure (LPP) and maximal bladder capacity (MBC) were quantified at the first week before transplantation and the 6th week after it.The expression of desmin,myosin heavy chain (MHC) and protein gene product 9.5 (PGP9.5) in the sphincter of rat urethra were analyzed,after the rats were sacrificed.Results The values of MBC and LPP were significantly higher than those before transplantation in HUMSCs treatment group (P < 0.01).In the HUMSCs treatment group,both values were significantly higher than those in the experimental control group,and the difference was statistically signifi cant (P < 0.01).Immunohistochemical staining and image analysis showed that the expression of desmin,MHC and PGP9.5 in the HUMSCs group was significantly higher than that of the experimental control group (P < 0.01).Conclusion HUMSCs transplantation for the treatment of SUI rats is effective and feasible.HUMSCs can improve uremic function by repairing the injured urethral sphincter and peripheral nerves.
