利用ISSR和叶绿体trnL-trnF序列变异研究葡萄种质资源的遗传多样性和系统发育关系

         

摘要

Nuclear ISSRs and cpDNA non-coding trnL-trnF region were used to analyse the genetic diversity and phylogenetic relationships of 36 grape accessions of Vitis vinifera L. Thirteen ISSR primers produced 63 bands in total,and the average values of the number of effective alleles,Nei’ s gene diversity,and Shannon’ s information index were 1. 5827,0. 3463,and 0. 5172,respectively. The ISSR UPGMA cluster analysis showed six major clusters. Vitis vinifera showed a high level of genetic diversity (Hd=0. 667,Pi=0. 00206) in the trnL-trnF region. The relationships inferred from the NJ,ML and BI analyses did not show geographical corre-spondence. Selective neutrality tests indicated that there had been a population expansion in their evolutionary history. In-situ conservation should be the most important approach. These results will be useful for the exploi-tation of grape germplasm in basic and applied research.%利用ISSR标记和叶绿体DNA trnL-trnF非编码区分析36种葡萄种质的遗传多样性和系统发育关系.13个ISSR引物共产生了63条带,平均有效等位基因数、Nei’s 基因多样性和香农信息指数分别是1.5827,0.3463和0.5172.基于UPGMA的ISSR分析将葡萄种质材料分为6个主要的进化枝.葡萄在trnL-trnF区域有较高的遗传多样性水平(Hd=0.667,Pi=0.00206).由NJ,ML和贝叶斯分析得到的进化树都没有表现出不同地区葡萄品种与地域之间的地理相关性.中性检验葡萄的进化历史过程中发生过种群扩张事件.葡萄种质资源的保护应以就地保护为主.本研究为葡萄种质资源的理论和应用研究提供了有价值的信息.
