百里醌抑制膀胱癌细胞生长的实验研究

         

摘要

目的 探讨百里醌对体外膀胱癌细胞株BIU -87生长的影响及其机制.方法 百里醌单独作用人膀胱癌细胞株BIU - 87及预处理NF - κB激活剂TPA后百里醌再作用BIU - 87细胞,四甲基偶氮唑蓝(MTT)法检测细胞增殖;流式细胞术检测细胞凋亡;Western blotting检测膀胱癌细胞质中IκBα蛋白表达.结果 与对照组相比较,百里醌可显著抑制体外膀胱癌细胞株BIU -87生长,并明显诱导细胞凋亡;预处理TPA可显著削弱百里醌对BIU -87细胞的增殖抑制和凋亡诱导作用;百里醌干预后膀胱癌细胞质中IκBα的表达明显上调.结论 百里醌可显著抑制体外膀胱癌细胞生长,并诱导细胞凋亡,该作用可能通过促进其胞质中IκBα的表达而实现.%Objective To investigate the effect and the mechanism of thymoquinone in the growth inhibition of bladder cancer in vitro. Methods After being treated with thymoquinone, or pretreated with NF - κB activator TPA, human bladder cancer cell line BIU -87 then got treatment of thymoquinone. The cellular proliferation was detected by methyl thiazolyl tetrazolium ( MTT) assay. The flow cy-tometry was used to determine apoptosis in bladder cancer cells. Western blotting was used to detect the expression of IkBcc in bladder cancer cells. Results The proliferation of bladder cancer cells was inhibited significantly by thymoquinone. Apoptosis rate induced by the thymoquinone was markedly higher than that of control. However, being pretreated with TPA could significantly attenuate the anti - proliferation and apoptosis induction effects of thymoquinone on the human bladder cell line BIU -87. The expression of IkBcc was up - regulated in BIU - 87 cells after treatment of thymoquinone. Conclusion Thymoquinone exerts anti - tumor activity in bladder cancer in vitro, which may be related to up - regulation of IkBcc.
