母鼠孕期和哺乳期暴露壬基酚对其仔鼠肾脏增殖细胞核抗原表达的影响

         

摘要

目的:探讨母鼠孕期和哺乳期暴露壬基酚(NP)对其仔鼠肾脏增殖细胞核抗原(PCNA)表达的影响。方法试验分为低、中、高3个剂量染毒组和1个对照组。母鼠受孕第6天到出生后第21天灌胃染毒NP,仔鼠出生60 d后剖杀取肾脏,免疫组化检测PCNA的表达。结果肾小管PCNA阳性细胞较肾小球多。与对照组比较,低、中、高剂量组仔鼠肾脏PCNA阳性细胞数明显增多,光密度增高,差异均有统计学意义(P<0.01),随着给药剂量的增加,PCNA阳性细胞数目逐渐增多,阳性细胞数与NP暴露剂量呈正相关(r=0.979,P<0.05)。结论母鼠孕期和哺乳期暴露NP使其仔鼠肾脏PCNA的表达增加。%Objective To investigate the influence of exposure to nonylphenol(NP) on proliferating cell nuclear antigen (PCNA) in the kidney of newborn rats during pregnancy and lactation. Methods The experiment was divided into 3 exposure groups(low dose group,medium dose group and high dose group) and control group. NP exposure time was limited from the 6th d after conception to 21st d after birth,the kidney was got after the newborn was killed 60 d later,and the expression of PCNA was detected by immunohistochemistry. Results The cells in renal tubular was more than those in renal glomerular. Compared with the control group,the number of positive cells in PCNA of rats′kidney increased obviously in low,medium and high dose group and the number of integrated optical density(IOD) also rose,the differences had statistically significant difference(P<0.01). With the increase of dosage of administration,the number of positive cells in PCNA rose gradually, and the NP exposure dose was as sociated with PCNA-positive cells in a dose-dependent manner(r=0.979,P<0.05). Conclusion Exposure to NP of female rats during gestation will induce an increase in PCNA expression in the kidney of newborn rats.
