紫苏密码子偏好性分析及fad3基因的异源表达预测

         

摘要

Perilla is one of the best oil crops with high level of α-linolenic acid (ALA).It is important to reveal the regulatory mechanisms of ALA biosynthesis for its industrial production,based on the codon usage bias and appropriate heterologous host of perilla genes.In this study,codon characteristics,codon numbers,relative synonymous codon usage,optimal codon,base correlation,parity rule and microbial heterologous expression of pfFAD3s were respectively analysed for gene using,according to the RNA sequence from seven development period of perilla seeds.Results showed that GC content was 45.85% in transcribed perilla sequences.Most gene codon bias were not obvious and tend to end with A/T base.Saccharomyces cerevisiae may be a more suitable host for pfFAD3s expression.This research provides a foundation for the synthesis regulation mechanism of perilla ALA and its large-scale production by microbial gene engineering.%紫苏α-亚麻酸(ALA)含量极高,其合成相关基因偏好性及异源表达宿主的阐明,是揭示紫苏不饱和脂肪酸调控机制及表达生产的重要前提.本研究从密码子特性、有效密码子数、相对使用度、最优密码子、碱基相关性、奇偶偏好性及异源表达特性等方面,对不同形成时期的紫苏种子转录组序列进行密码子偏好性分析,并对筛选出的pfFAD3s基因进行三种微生物异源表达预测分析比较.结果表明:紫苏转录核酸序列中的GC含量为45.85%,大部分基因密码子偏好性相对较弱,其蛋白编码基因密码子偏好以A/T结尾;pfFAD3s基因异源表达的适宜微生物宿主为酿酒酵母.分析结果为进一步研究紫苏ALA合成的分子调控奠定了基础,并为其微生物基因工程生产提供了一定依据.
