ET-1对高血压大鼠脑基底动脉血管平滑肌细胞增殖的作用

         

摘要

目的:观察内皮素-1(ET-1)对高血压大鼠脑基底动脉血管平滑肌细胞增殖的作用。方法将高血压大鼠和正常血压大鼠按加入 ET-1浓度的不同各分为6组:10%胎牛血清(对照组);10%胎牛血清+20μmol·L-1 ET-1(20μmol·L-1组);10%胎牛血清+40μmol·L-1 ET-1(40μmol·L-1组);10%胎牛血清+60μmol·L-1 ET-1(60μmol·L-1组);10%胎牛血清+80μmol·L-1 ET-1(80μmol·L-1组)及10%胎牛血清+100μmol·L-1 ET-1(100μmol·L-1组)。使用细胞计数试剂盒计数各组细胞个数;采用氚标胸腺嘧啶脱氧核苷([3 H]-TdR)掺入法测定各组血管平滑肌细胞的 DNA 的合成量作为细胞增殖的指标。结果ET-1刺激高血压大鼠脑基底动脉平滑肌细胞增生。高血压大鼠20、40、60、80及100μmol·L-1各组脑血管平滑肌细胞数量及 DNA 合成量较对照组均增加,以80 mol·L-1组细胞数量及 DNA 合成量增加最为明显(均 P <0.05)。正常血压鼠各组脑血管平滑肌细胞数量变化及 DNA 合成量增加不明显(P >0.05)。结论ET-1可明显促进高血压大鼠脑基底动脉血管平滑肌细胞增殖,提示 ET-1可能促进高血压诱发的脑血管重构。%ABSTRACT:Objective To investigate the effect of endothelin-1 (ET-1)on the proliferation of cerebral basilar artery smooth muscle cells in hypertensive rats.Methods The cerebral basilar ar-tery smooth muscle cells from hypertensive and normotensive rats were treated with ET-1(0,20, 40,60,80 or 100 μmol·L-1 )in the presence of 10% fetal bovine serum.Cell number was quanti-fied by using a Cell Counting Kit.Cell proliferation was evaluated by DNA synthesis,which was measured by 3H-TdR incorporation.Results ET-1 treatment stimulated the proliferation of cere-bral basilar artery smooth muscle cells in hypertensive rats.Compared with control group,the number of cerebral artery smooth muscle cells and the amount of DNA synthesized were signifi-cantly increased by treatment with 20 to 100 μmol·L-1 ET-1(P <0.05).The maximal effect was observed at the concentration of 80 μmol·L-1 .In contrast,ET-1 had no effect on the number of cerebral artery smooth muscle cells and the amount of DNA synthesized in normotensive rats(P >0.05).Conclusion The ET-1 can significantly stimulate the proliferation of cerebral basilar artery smooth muscle cells in hypertensive rats,suggesting that ET-1 may promote hypertension-induced cerebrovascular remodeling.
