肿瘤抗原 MAGE-A9和 MAGE-A11在乳腺癌中的表达及意义

         

摘要

目的::分析人黑色素瘤相关抗原MAGE-A9和MAGE-A11在乳腺正常组织、乳腺良性瘤及乳腺癌组织中的表达趋势,为乳腺癌临床诊断和生物治疗探寻新靶点。方法:选取本院接受并行手术治疗的乳腺良性肿瘤和乳腺癌患者各33例,分别取病灶组织和病灶周边正常组织制备石蜡病理切片,以免疫组织化学实验检测MAGE-A9和MAGE-A11的表达水平,并分析其与乳腺癌病理特征的相关性。结果: MAGE-A9在正常组织中无表达,在乳腺良性瘤和乳腺癌的阳性表达率分别为15.2%和54.5%,差异有统计学意义(P<0.05);MAGE-A11在正常组织及良性瘤中均无表达,仅在乳腺癌中表达,阳性表达率达78.8%。 MAGE-A9表达受组织学分级、ER、HER-2的影响,组织学Ⅲ级患者明显高于Ⅰ、Ⅱ级,ER、HER-2阳性高于阴性;MAGE-A11表达仅受到ER、HER-2的影响,ER、HER-2阳性高于阴性,差异有统计学意义(P<0.05)。线性回归分析结果显示MAGE-A9和MAGE-A11表达水平呈直线正相关性(y=0.7981x +1.278;R2=0.6948)。结论: MAGE-A9和MAGE-A11可能与乳腺癌的发生发展有密切关系。%Objective: To analyze the expression difference of melanoma associated antigen subtypes MAGE-A9 and MAGE-A11 in breast normal tissue,breast benign tumor and breast cancer,so as to search new targets for the clinical diagnosis and treatment of breast cancer. Methods:33 patients with breast normal tissue and 33 patients with breast cancer accepted in our hospital and treated with surgery were selected,and both the pathological tissue and the healthy tissue around were prepared paraffin slice. The MAGE-A9 and MAGE-A11 expression level were detected by immunohistochemical experiment,which relationship to pathological features were an-alyzed. Results:The MAGE-A9 had no expression in healthy tissue,and its expression rate in breast benign tumor and breast cancer ac-counted for 15. 2% and 54. 5%,which has statistical differences (P<0. 05). The MAGE-A11 had no expression in both healthy tissue and breast benign tumor,and its expression rate in breast cancer accounted for 78. 8%. The MAGE-A9 expression was affected by histo-logical grade and ER/HER-2,and those in GradeⅢand ER/HER-2 positive were higher that the others (P<0. 05). The MAGE-A11 expression was affected by ER/HER-2 only,and those in ER/HER-2 positive were higher than negative,the difference was statistically significant (P<0. 05). MAGE-A9 and MAGE-A11 is expected to become the new targets for clinical diagnosis of breast cancer and tumor immunotherapy. Linear regression analysis results showed that the MAGE-A9 and MAGE-A11 expression level had positive linear correlation (y=0. 798 1x+1. 278;R2 = 0. 694 8). Conclusion:MAGE-A9 and MAGE-A11 is expected to become the new targets for clinical diagnosis and tumor immunotherapy of breast cancer.
