X射线对仔鼠十二指肠组织结构和抗氧化酶活性的影响

     

摘要

为研究X射线辐射对仔鼠十二指肠组织结构和总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-PX)、谷胱甘肽还原酶(GR)活性的影响,用0,1,3,5和7 Gy的X射线对160例出生6~7 d的仔鼠进行全身辐射,分别于辐射后1,5,10和20 d,用比色法检测十二指肠中T-AOC,GSH-PX,GR的变化,用显微技术观察十二指肠组织结构的变化.结果表明,1 Gy组T-AOC和GR活性在辐射后1~20 d高于对照组,其他各组T-AOC和GR在辐射后始终低于对照组;1 Gy组GSH-PX活性在辐射后1 d低于对照组,后逐渐升高,并始终高于对照组;3 Gy组GSH-PX在辐射后1 d高于对照组,后降低,并低于对照组,其它各辐射组GSH-PX活性始终低于对照组;辐射组仔鼠十二指肠粘膜上皮和隐窝均有不同程度的损伤,3 Gy组在辐射后10 d损伤恢复,随着辐射剂量的增大,十二指肠粘膜上皮掉落、溶解,隐窝空泡化甚至消失,中央乳糜管扩张,小肠出血.表明X射线辐射影响仔鼠十二指肠组织结构,可能与十二指肠抗氧化酶活性的变化有关.%To explore the effects of X-ray on duodenum of filial mice, the changes of the histological structure, activities of T-AOC, GSH-PX and GR in duodenum are investigated after irradiation with different dosages of X-ray in development filial mice. 160 filial mice(birth 6~7 days) are irradiated with different dosages(0,1,3,5,7 Gy) X-ray, and then detecte the activities of T-AOC, GSH-PX, GR by colorimetry from all irradiated groups of different stages at 1 day, 5 days, 10 days and 20 days after irradiation. In addition, the changes of the duodenum lesions of filial mice are observed by optical microscope from all experimental groups. The intensities of T-AOC, GR activities in duodenum of the neonatal mice are lower in X-ray irradiated groups than that in the control group(P<0.05 or P<0. 01),with the exception for 1Gy group. The intensities of GSH-PX activities in duodenum of the neonatal mice are lower in 1Gy group and higher in 3Gy group than that in the control group on the first day after irradiation(P<0.05). And then, the activities of enzyme are increased in 1Gy group and reduced in 3Gy group at 5d~20d after irradiation, in other irradiated groups the intensities of GSH-PX are lower than that in the control group invariably(P<0.05 or P<0.01). With the increase of radiation dosages, the epithelial cells of duodenum and small intestinal crypts of filial mice have different degrees of change.Duodenum of 3. 0Gy experimental group in 10 days after radiation, injury recovery in general, in high dosages experimental groups, the epithelial cells fall-off and dissolve, small intestinal crypts vacuolization and disappear even, central lacteal expand extremely, hemorrhage of duodenum. X-ray radiation affects the structure of filial mice duodenum, it might be correlated with the low activities of T-AOC, GSH-PX and GR in filial mice duodenum.
