兔牙移动中半胱氨酸蛋白酶抑制剂C表达与牙周骨改建的研究

         

摘要

Objective:To invesligale the expression of cyslalin C during orlhodonlic tooth movement in rabbils. Methods, Experimental models of orlhodonlic loolh movement were established in 20 Japanese Big Ear Rabbils with about 2. 0 kg weighl. 0. 8 N orlhodonlic force was applied on the models. The gingival crevicular fluid was oblained and the conlenl of cyslalin C was measured by ELISA 1, 3, 5, 7 and 14 days after the force application, then, the animals (n =4) were sacrificed respectively. The morphology was observed by HE staining. The cystalin C expression in the bone was observed and quantified with SABC. Results, In the early stage (at 1 ~ 3 d) of tooth movement cystalin C expression decreased and osteoclasls increased, in the late stage (at 5 -14 d) the cystalin C expression increased and osteoclasls decreased. Conclusion, Cystalin C participates in proteolysis during orthodontic tooth movement.%目的:研究兔牙移动过程中半胱氨酸蛋白酶抑制剂C(cystatin C)在牙周组织中的表达变化,以及加力不同时间的变化特点.方法:选用20只2.0kg体重的日本大耳兔建立正畸牙移动模型,加力0.8N,分别在1、3、5、7、14d后取龈沟液,ELISA检测龈沟液中cystatin C的含量,取龈沟液后,处死动物,HE染色观察牙周组织改建的变化,免疫组化SABC法检测cystatin C的表达随着时间变化的特点.结果:cystatin C在正畸牙移动初期(1~3d)含量减少,破骨细胞增多,后期(5~14d)cystatin C含量增多,破骨细胞减少.结论:在正畸力的诱导下,cystatin C参与了正畸牙移动骨改建过程中有机基质的降解.
