三种钛金属表面处理方法对牙周膜成纤维细胞附着和生长的影响

         

摘要

目的:比较三种不同表面处理方法对牙周膜细胞在钛金属表面附着和生长影响。方法:将三种经不同方法表面处理的钛金属(纯钛、钛75、钛表面氮化处理)试件放在12孔培养板内,取生长良好的第5代人牙周膜成纤维细胞(PDLF)接种在试件表面,分别在接种后24 h和72 h进行贴壁细胞计数,并在扫描电镜下观察细胞在金属表面附着情况。结果:接种24 h 后纯钛表面的贴壁细胞数多于钛75金属表面的细胞数(P<0.05),但在接种后72 h时这种差异消失;不论接种后24 h或72 h钛表面氮化处理金属表面的细胞数都明显少于另外两种金属表面的细胞数(P<0.05)。扫描电镜结果表明PDLF在纯钛和钛75表面伸展良好,而在钛表面氮化处理的金属表面细胞基本不伸展。结论:人PDLF在纯钛、钛75表面的生物性附着优于在表面氮化处理的钛金属表面。%Objective: To compare the effect of three kinds of surface treatment on the attachment of periodontal ligament fibroblasts (PDLF) on titanium. Method:5×104 PDLFs in 100 μl of culture medium were seeded on the surfaces (1 cm2) of pure titanium,75-titanium, and nitrogenic-titanium respectively.The cells attached to the metal surfaces were trypsinized,suspended in culture medium and counted 24 h and 72 h after seeding respectively.Morphology of the cells attached to the metal surfaces was observed with scanning electron microscepe. Results:24 h after seeding the cells (1×104/ml) attatchad to pure titanium,75-titanium and nitrogenic-titanium were 5.1±0.9,2.6±0.4 and 0.2±0.1 respectively;while 72 h after seeding those were 12.8±0.8,10.3±0.8 and 3.3±0.3 respectively.Attached cells on pure titanium surface and 75-titanium surface were more outspread than those on nitrogenic-titanium surface. Conclusion:Pure titonium or 75-titanium suface may be more feasible for PDLF to attach and grow.
