诺丽内生真菌的分离鉴定及其次生代谢产物的抗氧化活性

         

摘要

Fruits and leaves of noni (Morinda citrifolia Linn.) were collected in Wanning,Danzhou,Qionghai,Sanya,Lingao and Haikou,Hainan Province,from which 32 strains of endophytic fungi were isolated,cultured at 25 ℃ (and) then identified by using 26S rDNA D1/D2 and ITS rDNA sequencing.The results showed that these 32 strains belonged to the phylum Basidiomycota and the phylum Ascomycota.Of the 32 strains 13 belonged to 3 genera,i.e.Aureobasidium,Rhodotorula and Cryptococcus,and 5 species,and 19 belonged to 7 genera,i.e.Colletotrichum,Diaporthe,Hypoxylon,Cladosporium,Fusarium,Phyllosticta and Gibberella,and 10 species.The secondary metabolites were extracted with ethyl acetate as solvent from 15 fermented solutions of noni endophytic fungi,and their antioxidant activity was evaluated with ABTS +·-free radical scavenging assay.The total polyphenols content (TPC) and total flavonoid content (TFC) of the secondary metabolites were determined by using Folin-reagent method and A1Cl3 coloritnetric method.The antioxidant assay showed that ethyl acetate extract from 15 solutions of endophytic fungi was full of antioxidant activity,and that the antioxidant activity of M48 and J22 was significantly different.Noni endophytic fungi had abundant diversity and the potential to produce antioxidant substances which have important application value and broad prospect in healthcare and medical sector.%对采自海南省海口市、万宁市、儋州市、琼海市、三亚市和临高县的诺丽果和叶,消毒后,25℃下分离培养得到32株内生真菌,采用26s rDNA D1/D2区域和ITS rDNA测序鉴定菌种.结果表明:32株内生真菌属于担子菌门(Basidiomycota)、子囊菌门(Ascomycota)2个类群.其中,13株酵母菌分别属于金担子菌属(Aureobasidium)、蔷薇色酵母属(Rhodotorula)、隐球菌属(Cryptococcus)3个属5个种;19株霉菌分别属于刺盘孢属(Colletotrichum)、间座壳属(Diaporthe)、炭团菌属(Hypoxylon)、枝孢属(Cladosporium)、镰刀菌属(Fusari-um)、叶点霉属(Phyllosticta)和赤霉属(Gibberella)7个属10种.同时,采用乙酸乙酯溶剂对15种诺丽内生真菌发酵液进行提取,获得次生代谢产物.用ABTS+·自由基清除法进行了抗氧化活性评价,用福林-肖卡法和AlCl3显色法测定各提取次生代谢产物中总多酚含量(TPC)、总黄酮含量(TFC).结果显示,乙酸乙酯提取的15种诺丽内生真菌都具有抗氧化活性,且m48,j22抗氧化活性显著,诺丽内生真菌具有较丰富的多样性和具有产生抗氧化活性物质的潜力,在医药健康领域具有重要应用价值.
