低热河谷区薏苡仁油的提取及对癌细胞的抑制研究

         

摘要

以地热河谷地区的薏苡仁为研究对象,提取薏苡仁油并进行抗肿瘤活性测定,目的是为提高薏苡仁油的得率,为工业化提取薏苡仁油提供参考.以提取时间(30-50min)、 提取温度(40-60℃)、 料液比(1︰2.5-1︰4.5m/v)和超声功率(100-140W)为因素,采用L9(34)正交试验优化了最佳工艺,并进一步进行薏苡仁油对肿瘤细胞的抑制活性研究.结果表明,上述4个因素对高良乡薏苡仁油提取率均有一定的影响,提取时间、提取温度、 料液比和超声功率对薏苡仁油提取率的贡献率分别是74.47%、16.53%、5.11%和2.83%,并通过直观分析和方差分析,得知薏苡仁油的最佳提取工艺为以无水乙醇为溶剂,料液比1︰2.5 m/v、 提取温度60℃、 提取时间50min、 超声功率100W,在此提取条件下薏苡仁油的提取率为7.68±0.12%,采用该提取工艺得到的薏苡仁油对结肠癌细胞(SW480)、 人乳腺癌细胞(MCF7)、 人肝癌细胞(SMMC7721)、 人肺癌细胞(A549)、 人早幼粒白血病细胞(HL-60)5种肿瘤细胞的抑制率分别为95.52%、85.02%、91.83%、92.54%、72.89%.该研究结果为将薏苡仁油开发成抗肿瘤保健植物油奠定理论基础.%In order to improve extraction yield of coix-see from low-hot valley was targeted as research object, coix-seed oil was extracted and its antineoplasmic activity were studied in this paper.Methods for improving extraction yield of coix-seed oil were optimized through L9 (34 ) or-thogonal experiments with the factors including extraction time (30-50min) , extraction temperature (40-60℃) , ratio of material to liquid (1 ︰2.5-1 ︰4.5m/v), ultrasonic power (100-140W).Furthermore, inhibiting effect of coix-seed oil on some cancer cells was tested.The results showed that yield of oil from seed of coix was significantly affected by four factors, contribution rate of extraction time, extraction temperature, ratio of material to liquid and ultrasonic power, which were 74.47%, 16.53%, 5.11%, 2.83%, respectively.The optimized extraction method was confirmed based on visual and variance analysis that absolute ethyl alcohol was considered as optimal extraction solvent, with optimal extraction time, extraction temperature, ratio of material to liquid and ul-d oil and provide a reference for industrialization, coix trasonic power of 50 min, 60℃, 1︰2.5m/v, 100W, respectively.Under these conditions, extraction yield of oil from seed of coix was 7.68 ±0.12%.And collected oil from coix seed could prevent growth of colon cancer cell (SW480), human breast cancer cell (MCF7), hepatoma carcinoma cell (SMMC7721), human lung cancer cell ( A549 ) , and leukemia cell ( HL-60 ) at inhibiting rate of 95.52%, 85.02%, 91.83%, 92.54%, 72.89%, respectively.The results of the study may lay a theoretical foundation for developing coix-seed oil as health-care veg-etable oil with antineoplastic activity.
