聚乙二醇沉淀猪胰蛋白酶的研究

         

摘要

To study the condition for extractaction of porcine trypsin by PEG (polyethyleneglycol) ,the effect of molecular weight and concentration of PEG, pH and extraction time on trypsin activity was investigated by both single factor experiment and orthogonal factor experiment. The optimum technology was determined by experiment as follows; 0. 25% of PEG 6000 with pH equal to 7. 5 and temperature of 4 ℃ while the extraction time was 3 h. Under the optimum conditions, the activity of the trypsin reached above 1600 u/ mL, higher than that of traditional ammonium sulphate precipitation. This method is effective to isolate trypsin from porcine pancreas.%研究了从猪胰脏中用聚乙二醇(PEG)分离提取胰蛋白酶的条件.采用单因素实验考查了聚乙二醇分子量、聚乙二醇浓度、pH值和提取时间对胰蛋白酶活力的影响,并通过正交实验对提取工艺条件进行优化,在0.25g/mL聚乙二醇6000,pH7.5,4℃提取3h条件下,胰蛋白酶酶活力达到1600 U/mL以上,比传统的盐析法提取率高,并且操作简单,是一种比较有效的分离猪胰蛋白酶的方法.
