基于NMR代谢组学方法研究异常黑胆质成熟剂对异常黑胆质型2型糖尿病大鼠的作用机制

         

摘要

Objective To study the mechanism of effect of Abnormal Savda Munziq to the Abnormal Savda Syndrome type 2 diabets rats .Methods Nuclear magnetic resonance (NMR) technology combined with pattern recognition and Orthogonal Projections to Latent Structures Discriminant Analysis (OPLS‐DA ) method were used to find out the variability urine metabolits of Abnormal Savda Syndrome type 2 diabets rats after oral administration of low ,medium ,high dose of Abnormal Savda Munziq (ASM ) for 21 days . Results After oral administration of ASM ,the reduced concentration level of some metabolits including leucine ,isoleucine ,Phenylalanine ,critic acid ,α‐oxoglutaric acid ,hippuric acid and formic acid were in‐creased in Abnormal Savda Syndrome type 2 diabets rats′urine ,together with the decreased concentration level of β‐glucose and α‐Glucose in Abnormal Savda Syndrome type 2 diabets rats′urine ,compared with Abnormal Savda Syndrome type 2 diabets rats group ,there were significant differences (P <0 .05) .Con‐clusion ASM has significant effects to the Abnormal Savda Syndrome type 2 diabets rats′urine metabo‐lism .It′s mechanism of effect may be achieved by readjusting the altered energy metabolism ,amino acid metabolism caused by Abnormal Savda Syndrome type 2 diabets .%目的:探讨异常黑胆质成熟剂(ASM )对异常黑胆质型2型糖尿病(T2DM )大鼠的作用机制。方法采用核磁共振(NMR)技术结合模式识别和正交偏最小二乘判别(OPLS‐DA)分析方法,寻找灌胃不同剂量ASM混悬液后异常黑胆质型 T2DM 大鼠尿液中的差异性代谢产物以及发生改变的代谢网络,分析ASM 对异常黑胆质型T2DM 大鼠尿液代谢的影响。结果灌胃ASM 后,在异常黑胆质型 T2DM 大鼠尿液中下降的亮氨酸、苯丙氨酸、异亮氨酸以及柠檬酸、α‐酮戊二酸、马尿酸、甲酸显著增加,同时在异常黑胆质型 T2DM 大鼠尿液中增多的β‐葡萄糖和α‐葡萄糖含量明显降低,与异常黑胆质型 T2DM 模型组比较差异有统计学意义(P <0.05)。结论 ASM对异常黑胆质型 T2DM 大鼠尿液代谢有显著影响,其作用机制可能是通过作用于异常黑胆质型T2DM引起的大鼠体内发生紊乱的糖代谢及氨基酸代谢而发挥治疗和预防异常黑胆质型T2DM的作用。
