不同频率电针对神经病理痛模型大鼠镇痛效应观察

         

摘要

[目的]观察不同频率电针对神经病理痛大鼠镇痛效应的影响。[方法]SD大鼠随机分为正常组、假手术组、模型组、2Hz电针组、15Hz电针组、50Hz电针组、100Hz电针组与120Hz电针组,每组6只。模型组采用脊神结扎(spinal nerve ligation, SNL)的方法制作大鼠神经病理痛模型。电针组采用电刺激器输出不同刺激参数,取大鼠患侧“足三里”“、昆仑”穴,于造模后第4天开始治疗,隔日1次,治疗5次。分别于造模前,造模后第3、4、6、8、10、12d,共7个时间点检测大鼠右后机械缩足阈( Paw Withdrawal Thresholds,PWTs)。[结果]造模后第4天,120Hz电针组大鼠痛阈高于模型组( P=0.044);造模后第6天,100Hz电针组和120Hz电针组大鼠痛阈均高于模型组( P=0.018和P=0.023);造模后第10天,2Hz电针组大鼠痛阈均高于模型组、15Hz电针组和50Hz电针组( P=0.024、P=0.036和P=0.002);造模后第12天,2Hz电针组、100Hz电针组和120Hz电针组大鼠痛阈均高于模型组( P=0.019、P=0.022和P=0.028)和50Hz电针组(均P=0.002)。[结论]在神经病理痛的早期,100Hz或120Hz频率电针的效果较2Hz频率、15Hz频率、50Hz频率的电针为佳;在神经病理痛慢性期,2Hz频率电针的效果较15Hz频率、50Hz、100Hz、120Hz频率频率的电针为佳。在电针镇痛过程中,不建议选用15Hz频率、50Hz频率的电针。%Objective] To investigate the analgesia effect of electro-acupuncture with different frequencies on neuropathic pain rats. [Methosds] SD rats were randomly divided into normal, sham spinal nerve ligation, SNL group, 2Hz EA group, 15Hz EA group, 50Hz EA group, 100Hz EA group, and 120Hz EA group, 6 animals per group. Rat neuropathic pain model was made by right L5 spinal nerve ligation in SNL group. EA groups were administered at right points Zusanli and Kunlun with different stimulation parameters through electrical stimulator from the 4th day after SNL for 5 treatments every other day. Before SNL operation, D3, D4, D6, D8, D10, D12 after operation PWTs were measured. [Result] D4 after SNL, 120Hz EA group PWTs is higher than SNL group( P=0.044) . D6 after SNL, 100Hz and 120Hz EA groups PWTs is higher than SNL group( P=0.018 and P=0.023) . D10 after SNL, 2Hz EA group PWTs is higher than SNL group, 15Hz EA group and 50Hz EA groups( P=0.024, P=0.036 and P=0.002) . D12 after SNL,2Hz, 100Hz and 120Hz EA groups PWTs is higher than SNL group( P=0.019, P=0.022 and P=0.028) and 50Hz EA group( all P=0.002) . [Conclusion] 100Hz or 120Hz EA is advised using in the early stages of neuropathic pain, and 2Hz EA in chronic neuropathic pain. We do not recommend for applicating 15Hz and 50Hz EA for analgesia.
