青蒿琥酯对人结肠癌细胞HCT116和HT29的增殖与凋亡

         

摘要

Objective:To study the effects of artemisine on proliferation and apoptosis of human colon cancer HCT116 and HT29 cells.Method:Human colon cancer cells were treated with artemisine at different concentrations. MTT assay was used to test cell proliferation and FCM was used to detect cell apoptosis.Result:The inhibition rates had upwarded trend with increasing concentrations (HCT116:19.42%,24.59%,44.84%,74.15%,92.87%;HT29:11.77%,17.63%,34.23%,48.51%,70.59%), when treated with artemisine at difierent concentrations (7.5, 15, 30, 60, 120 μg/mL) for 48 hours.Apoptosis rates gradually increased with increasing concentration (HCT116:2.46%, 51.36%,64.10%;HT29:1.14%,35.95%,43.45%), when treated with artemisine at different concentrations (0, 60, 120 μg/mL) for 24 hours.Conclusion:Artemisine inhibits proliferation and induces apoptosis of human colon cancer cells HCT116 and HT29.%目的:观察青蒿琥酯对人结肠癌细胞HCT116和HT29细胞增殖和凋亡的影响。方法:采用不同浓度青蒿琥酯处理HCT116和HT29细胞,采用四甲基偶氮唑蓝(MTT)比色法检测青蒿琥酯对人结肠癌细胞的增殖抑制效应,采用流式细胞术检测细胞凋亡的情况。结果:不同浓度青蒿琥酯(7.5、15、30、60、120μg/mL)处理两种人结肠癌细胞48 h后,抑制率随浓度增加呈上升趋势(HCT116:19.42%、24.59%、44.84%、74.15%、92.87%;HT29:11.77%、17.63%、34.23%、48.51%、70.59%)。不同浓度青蒿琥酯(0、60、120μg/mL)处理两种人结肠癌细胞24 h后凋亡率随浓度增加逐渐上升(HCT116:2.46%、51.36%、64.10%;HT29:1.14%、35.95%、43.45%)。结论:青蒿琥酯对人结肠癌CHT116和HT29细胞具有生长抑制和诱导凋亡作用。
