库存血加热对于红细胞免疫功能的影响

         

摘要

目的 通过检测37℃温浴前后正常献血员的红细胞上白细胞分化抗原相关指标来评价温浴对红细胞免疫功能是否有影响,并进一步探讨温浴不同温度和时间对红细胞CD35[红细胞补体受体1(CR1)]、CD59、CD55、CD58、CD44s表达的影响,为库存红细胞安全、有效地使用提供理论依据.方法 随机抽取河北省血液中心采集的无偿献血、经检测各项指标全部合格血液标本50份,每份均分成8段(A、B、C、D、E、F、G、H组)分别行4℃保存,37℃加热10,20,30,60,120,180 min和40℃加热30 min处理,并进行后续实验.采用流式细胞技术检测CR1及红细胞CD58、CD59、CD55、CD44s分子水平.结果 红细胞CD35、CD58、CD55、CD44s阳性率和分子水平在37℃加热后30 min内无显著变化,之后随着时间延长而逐渐降低.红细胞CD59阳性率和分子水平在37℃加热180 min内变化不显著.结论 库存红细胞37℃温浴30 min以内对红细胞免疫功能无明显影响,患者输注红细胞前库存红细胞37℃温浴30 min为最佳.%Objective It is to evaluate whether warm bath has effect on erythrocyte immune function by detecting correlated indexes of leukocyte differentiation antigens on erythrocyte in blood sample of healthy blood donors before and after 37 ℃ warm bath,and further more to confirm the effect of water bath at various times and different degrees on erythrocyte of CD35 [Erythrocyte complement receptor type 1 (CR1 0],CD59,CD55,CD58,CD44s expressions,thus to provide theoretical basis for safe and effective use of storing suspending erythrocytes.Methods A total of 50 whole blood specimen whose each index all qualified from Blood Center of Hebei Province were randomly selected.The tubes were sealed into 8 segments(A,B,C,D,E,F,G,H group) and were stored at 4 ℃,and then warmed at 37 ℃ for various time(10,20,30,60,120,180 min) and at 40 ℃ for 30 min,and then the test was continued.The levels of CR1,CD58,CD59,CD55 and CD44s were detected by flow cytometry technology.Results The molecular level and positive rate of CD35,CD58,CD55 and CD44s on red blood cells after water bath at 37 ℃ for 30 minutes did not significantly change,and then gradually decreased with time extended.The molecular level and positive rate of CD59 of red blood cells at 37 ℃ for 180 minutes did not significantly change.Conclusion Erythrocyte immune functions has not been significantly affected,if red blood cells are water bathed in 37 ℃ within 30minutes.It is optimal that storing suspending erythrocytes in warm bath at 37 ℃ for 30 minutes before transfused to patients.
