头花蓼治疗变应性接触性皮炎小鼠的疗效及作用机制探讨

         

摘要

目的:探讨头花蓼治疗变应性接触性皮炎( ACD)小鼠的疗效及机制。方法将36只Balb/C小鼠随机分成正常组、模型组、头花蓼低浓度组、头花蓼中浓度组、头花蓼高浓度组及地塞米松组6组,每组6只。除正常组外,其余组均采用异硫氰酸荧光素( FITC)诱导小鼠 ACD模型。造模成功第2天后,头花蓼低、中、高浓度组分别灌胃2.5%,5%,10%头花蓼溶液0.4 mL,地塞米松组灌胃0.4 mL地塞米松溶液,均连续7 d。观察各组小鼠皮损形态、组织病理改变以及血清白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)和肿瘤坏死因子-α(TNF-α)水平变化。结果治疗7d后,头花蓼低、中、高浓度组和地塞米松组小鼠的皮损形态和组织病理结构较治疗前明显改善;与模型组比较,头花蓼低、中、高浓度组和地塞米松组右耳厚度、中性粒细胞数、单核细胞数、血清 IL-4和 TNF-α水平均显著降低( P 均﹤0.05),但血清 IFN -γ水平比较差异均无统计意义(P均﹥0.05)。结论头花蓼对 ACD小鼠有显著治疗作用,可能与降低 IL-4和 TNF-α水平有关。%Objective It is to observe the effect and mechanism of Polygonum capitatum on allergic contact dermatitis ( ACD)of mouse model. Methods Thirty-six BALB/c mice were randomly divided into normal group,model group,Polygo-num capitatum low does group,middle does group and high does group,and dexamethasone group,6 cases in each group. Ex-cept the normal group,the ACD models were induced on the abdomen and ear with Fluorescein isothiocyanate in the other groups. The second day of successful model,the animals in Polygonum capitatum low does group,middle does group and high dose group were intragastriced administration with 2. 5%,5% and 10% Polygonum capitatum solution for 0. 4mL respectively, and with 0. 4mL dexamethasone in dexamethasone group,treatment for 7 days. The effect and mechanism of Polygonum capita-tum were evaluated by form lesions,pathological changes in tissue and the levels of serum interleukin-4(IL-4),interferon -γ( IFN-γ)and tumor necrosis factor( TNF-α)in mice. Results After seven days of treatment,the lesions form and his-topathological of Polygonum capitatum low does group,middle does group and high does group,and dexamethasone group were improved significantly compared with that before treatment. Compared with the model group,the right ear thickness,neutro-phil,monocytes,serum IL-4 and TNF-α were significantly lower in groups of Polygonum capitatum low does,middle does, high does group and dexamethasone group(all P ﹤0. 05),but there was no significance difference in serum IFN -γ levels ( all P﹥0. 05). Conclusion Polygonum capitatum has significant therapeutic effect on ACD,this may be related to lower the levels of IL-4 and TNF-α.
