超高效液相色谱-四级杆-飞行时间串联质谱法同时快速鉴定复方延寄参胶囊的化学成分

         

摘要

Objective To determinate the chemical components of Compound Yanjishen Capsules′methanol extracts simultaneously and rapidly by UPLC-Q-TOF-MS in negative ion mode.Methods The methanol extracts of Compound Yanjishen Capusles were prepared by ultrasonic extraction.The study was performed on an ACQUITY UPLCTMBEH C18column(100 mm×2.1 mm,1.7 μm)with acetonitrile-1mL·L -1formic acid in water as the mobile phase.The main components were separated by gradient elu-tion.The MS detection was performed with ESI source in negative ion mode.The main peaks were assigned.Results In Compound Yanjishen Capsules′methanol extracts,10 ginsenosides were qualitatively identified,including ginsenosides R1,ginsenosides Re, ginsenosides Rf,ginsenosides Rg1,ginsenosides Rb1,ginsenosides Rd,ginsenosides Ro,ginsenosides Rb2,ginsenosides Ra1and gin-senosides Rg3.3 flavonoids were identified,including neoeriocitrin,naringenin and naringin.3 phenolic acids were identified,inclu-ding fumaric acid,protocatechuic acid and caffeic acid.Conclusion In the study,16 compounds were identified under the same condi-tions by UPLC-Q-TOF-MS.The method has the advantages of high resolution,high efficiency and convenience,and can be used for the chemical components identification of Compound Yanjishen Capsules.%目的 采用超高效液相色谱-四级杆-飞行时间串联质谱(UPLC-Q-TOF-MS)法在负离子模式下对复方延寄参胶囊甲醇提取物的主要化学成分进行快速鉴定.方法 超声提取法制备复方延寄参胶囊甲醇提取物,采用ACQUITY UPLCTMBEH C18 (100 mm×2.1 mm,1.7 μm)色谱柱为分析柱,以乙腈-1 mL·L -1甲酸水为流动相,进行梯度洗脱分离各主要成分;质谱采用电喷雾离子源,负离子模式下扫描采集数据,对各主要色谱峰进行归属.结果 复方延寄参胶囊甲醇提取物中共定性鉴别出以下成分:人参皂苷类化学成分10种,包括三七皂苷R1、人参皂苷Re、人参皂苷Rf、人参皂苷Rg1、人参皂苷Rb1、人参皂苷Rd、人参皂苷Ro、人参皂苷Rb2、人参皂苷Ra1和人参皂苷Rg3;黄酮类化学成分3种,包括新北美圣草苷、柚皮素和柚皮苷;酚酸类化学成分3种,包括富马酸、原儿茶酸和咖啡酸.结论 采用UPLC-Q-TOF-MS法在同一条件下共定性鉴别出16种化合物,该方法分辨率高、高效、便捷,可快速准确地鉴别复方延寄参胶囊的化学成分.
