红花清肝十三味对CCl4诱导大鼠肝损伤纤维化的作用

         

摘要

目的 研究红花清肝十三味(HHQG)对CCl4诱导大鼠肝损伤纤维化的防治作用.方法 采用大鼠CCl4肝损伤模型,观察HHQG对肝损伤大鼠肝体指数及肝组织病理变化的影响,检测大鼠血清中的AST、ALT和LDH与肝组织匀浆中SOD、GSH、MDA、MMP-1和TIMP-1含量变化.结果 HHQG高剂量组肝体指数较模型组明显降低(P<0.05),肝组织炎症变性坏死及纤维组织增生较模型组大鼠明显降低,血清AST、ALT和LDH含量较模型组大鼠均明显降低(P<0.05);HHQG高剂量组肝匀浆中SOD和GSH活性较模型组明显升高,MDA含量明显降低;HHQG高剂量组肝匀浆中 MMP-1活性较模型组明显升高(P<0.05),TIMP-1活性明显降低(P<0.05)、MMP-1/TIMP-1比值较模型组提高66.6%.结论 HHQG对CCl4引起的大鼠肝损伤及纤维化有较好的防治作用,其机制可能与抗氧化作用调节MMP-1和TIMP-1细胞因子活性有关.%Objective To study the effect and mechanism of honghuaqinggan-13(HHQG)on liver injury induced by CCl4in rats. Methods HHQG on rat liver index and liver tissue pathologic changes and the effect on the serum AST,ALT,LDH and SOD, GSH,MDA,MMP-1 and TIPM-1 in liver tissue homogenate variation were observed by using CCl4hepatic injury model of rats. Results Liver volume index of HHQG high dose group was significantly lower than of the model group(P<0.05),Inflammation of the liver tissue degeneration necrosis and fibrous tissue hyperplasia of HHQG high dose group was significantly lower than of the model group(P<0.05).The serum AST,ALT,LDH content of HHQG high dose group were significantly lower than of the mod-el group(P<0.05).SOD and GSH activity significantly increased,MDA content decreased obviously,MMP-1 activity increased significantly(P<0.05),TIMP-1 activity significantly decreased(P< 0.05),and MMP-1/TIMP-1 ratio increased by 66.6% when compared with the model group.Conclusion HHQG has certain control effect on liver injury and fibrosis of rats caused by CCl4,its mechanism may be related to antioxidant effect on regulating the activity of MMP-1 and TIMP-1.
