Yeast paste is one of the main byproducts of beer fermentation process,accounting for about 1.5% of the beer production,which is rich in glutathione (GSH).Ethanol extraction was used to improve the extraction efficiency of GSH from yeast paste.Response surface methodology (RSM) was used to optimize the glutathione extraction process using the ethanol from fresh yeast paste.According to Box-Behnken design principle based on single-factor experiments,the extraction process of Glutathione was optimized.The method includes response surface analysis with three factors and three levels combined with the Design-Expert 8.0.6 data analysis software.It was adopted to study three factors including ethanol volume fraction,extraction temperature and extraction time,and the effect of their interactions on the extraction process.The results showed that the Glutathione yield reached 0.258% when the ethanol volume fraction,extraction temperature and extraction time was 30.98%,35.81℃,and 59.39 min,respectively.The final yield of Glutathione under this optimum preparation conditions was almost equal to the actual measured value.Ethanol extraction is superior to hot-water extraction methods.Therefore,the obtained extraction conditions are very valuable to.practical applications.%酵母泥是啤酒发酵过程中的主要副产物之一,约占啤酒产量的1.5%,啤酒废酵母中谷胱甘肽(GSH)含量较高.为提高酵母泥中GSH提取效率,采用乙醇提取法,并通过响应面法对乙醇提取新鲜酵母泥GSH工艺条件进行优化,在单因素实验的基础上,根据Box-Behnken实验设计原理,运用Design-Expert 8.0.6数据统计分析软件,采用三因素三水平的响应面分析法,研究乙醇体积分数、提取温度和提取时间3个因素及其交互作用对提取工艺的影响.确定最优工艺参数是:乙醇体积分数30.98%,提取温度35.81℃,提取时间59.39 min,GSH理论得率0.258%.与优化后的实际得率非常接近,与传统热水提法(0.168%)相比有明显优势,得到的GSH提取条件具有实际应用价值.
