目的 探讨峨参内酯联合5-FU对CT26结肠癌皮下移植瘤生长的影响.方法 借助鼠源结肠癌细胞CT26构建Balb/c小鼠移植瘤模型,将接种成功后的小鼠随机划分为5组,即空白组、模型组、峨参内酯组、5-FU组、5-FU+峨参内酯组,其中前两组为对照组,分别给予0.5％CMC-Na和生理盐水,后三组为实验组,分别进行不同的药物治疗.通过小鼠一般状态、脾脏、胸腺指数、对药物抑制率等指标衡量治疗情况,借助ELISA检测不同小组血清中COX2、iNOS和NF-κB具体情况.结果 峨参内酯对Balb/c小鼠CT26结肠癌皮下移植瘤抑瘤率为18.81％;单独应用5-FU的抑瘤率为76.60％,联合应用峨参内酯和5-FU的抑瘤率为89.44％;峨参内酯有助于改善小鼠免疫能力,可减少小鼠血清中COX2、iNOS、NF-κB的含量.结论 峨参内酯可有效抑制Balb/c小鼠CT26结肠癌皮下移植肿瘤发生几率,联合5-FU可有效缓解毒性,提高功效.%Objective To inverstigate the effects and mechanisms of Anthricin combined with 5-FU for the growth of colon carcinoma in mice implanted with CT-26 cell.Methods The mouse transplanted colon cancer cell CT26 is used to construct the BALB model,and the mice are randomly divided into 5 groups,namely,blank group,model group,Anthricin group,5-FU Group,5-FU+ Anthricin lactone group.The first two groups are the control group,the treatment is 0.5 ％ CMC-Na and saline,and the last three groups are treated with different drugs.The specific concentrations of COX2,Inos and NF-κB contained in different groups of serum are detected by Elisa in the general state,spleen,thymus index and drug inhibition rate.Results Anthricin on Balb/c mice CT26 colon cancer xenograft tumor inhibition rate is 18.81％;the single application of 5-FU inhibition rate is 76.60 ％,the combined application of anthricin and 5-FU inhibition rate is 89.44 ％;The content of COX2,iNOS and NF-kB in mice serum can be reduced by Anthricin,which can improve the immune ability of mice.Conclusion Anthricin can effectively inhibit the incidence of CT26 colorectal cancer in Balb/c mice,combined with 5-FU can effectively alleviate toxicity and improve efficacy.
