目的 以胡椒碱为母核进行结构修饰,观察胡椒碱及其衍生物5-(3’,4’-次甲二氧苯基)-戊烯酰丙胺的抗抑郁活性.方法 采用小鼠强制游泳实验、悬尾实验,观察胡椒碱和5-(3’,4’-次甲二氧苯基)-戊烯酰丙胺对小鼠悬尾不动时间、游泳不动时间的影响.结果 胡椒碱(10,20mg·kg-1)和其衍生物5-(3’,4′-次甲二氧苯基)-戊烯酰丙胺(10,20mg·kg-1)给药后,均能明显缩短小鼠在行为学实验中的不动状态时间(P<0.01),且5-(3’,4’-次甲二氧苯基)-戊烯酰丙胺较胡椒碱更为明显.结论 胡椒碱和5-(3’,4′-次甲二氧苯基)-戊烯酰丙胺具有明显的抗抑郁作用,且5-(3’,4’-次甲二氧苯基)-戊烯酰丙胺的抗抑郁作用优于胡椒碱.%Objective To observe the antidepressant effect of piperine and its derivative 5-(3', 4'-methylenedioxyphenyl)-2E,4E-pentadienoic acid n-propylamine amide (MDPP), Piperine was modified. Methods Mouse tail suspension test (TST) and mouse forced swimming test (FST) were adopt to observe the effect of piperine and MDPP in depression . Results Piperine (10, 20 mg·kg-1) and MDPP (10, 20 mg·kg-1) were both able to elicit time-dependent reduction in immobility duration in TST and FST apparently (P<0.01), but MDPP (10, 20 mg·kg-1) were more effective. Conclusion Piperine and MDPP had antidepressant effect, and MDPP was more significant.
