PSF1在肝细胞肝癌组织中的表达及其临床意义

         

摘要

为探讨肝细胞肝癌中PSF1的表达情况及其临床意义,利用免疫组化技术检测PSF1在肝细胞肝癌及其对应癌旁组织中的表达水平,并运用SPSS17.0软件分析PSF1与肝癌临床病理特征之间的关系.在肝癌细胞系MHCC97-H、HepG2、SMMC-7721和肝细胞系HL-7702中运用实时定量PCR检测PSF1的表达情况.在肝癌细胞系MHCC-97H中,通过siRNA特异性下调PSF1的表达,通过Transwell小室实验检测PSF1对MHCC-97H细胞侵袭能力的影响.所得结果表明:与癌旁组织相比,PSF1在90对肝癌癌旁组织中有67.7% (60/90)的样本高表达(x2=28.611,p<0.01).统计分析发现PSF1高表达与患者有无肝硬化(x2=4.703,p<0.05)、是否发生远处转移及门脉侵犯(x2=6.939,p<0.05)、肿瘤分化程度(x2=4.293,p<0.05)、TNM分期(x2=16.987,p<0.01)显著相关;其表达水平与患者性别、年龄、肿瘤大小、肿瘤数目无关.在肝癌细胞系MHCC97-H、HepG2、SMMC-7721和肝细胞系HL-7702中,其表达逐渐降低,提示PSF1可能参与了肝癌的侵袭进程.在MHCC97-H中采用siRNA技术特异性下调PSF1的表达后,其侵袭能力降低.PSF1在肝癌组织中高表达且与患者远处转移及门脉侵犯、TNM分期相关,抑制PSF1的表达后MHCC-97H侵袭能力减弱,PSF1可能作为治疗肝癌的一个靶点.%In order to investigated the expression of PSF1 in HCC and its adjacent benign tissues and evaluated the relationship between PSF1 with clinicopathological features of HCC, the expression of PSF1 in HCC and its adjacent benign tissues by Immunohistochemistry and analysed its relationship with clinicopathological features of HCC were investigated by SPSS17.0.The expression of PSF1 was analyzed by RT-PCR in cell lines (MHCC97-H, HepG2, SMMC-7721, HL-7702).PSF1 was down-regulated by siRNA interference in MHCC97-H, the invasion capacity of HCC cells was detected with Transwell cell culture chambers.The results show that PSF1 expression is up-regulated in 67.7% (60/90) of HCC tissues compared with adjacent benign tissues.Statistical analyses reveale that high PSF1 expression levels are significantly associated with liver cirrhosis (x2=4.703,p<0.05), microvascular invasion or metastasis (x2=6.939,p<0.05), tumor differentiation (x2=4.293,p<0.05), and TNM stage (x2=16.987,p<0.01).But there was no significance with respect to gender, age, tumor number, tumor size.The expression of PSF1 in HCC cell lines (MHCC97-H、HepG2、SMMC-7721) and in a human liver non-tumor cell line (HL-7702) was evaluated.This result showed that the expression of PSF1 decreases gradually in the above cell lines, which suggests that PSF1 may participate in the cellular invasion process of HCC cells.Transfection of MHCC97-H cells with PSF1-siRNA, down-regulated PSF1 could inhibit the invasion of HCC cells.High expression of PSF1 was observed in 67.7% (60/90) of HCC tissues compared with adjacent benign tissues and high PSF1 expression levels were significantly associated with microvascular invasion or metastasis, TNM stage.Transfection of MHCC97-H cells with PSF1-siRNA, the down-regulation of PSF1 expression decreased the invasion capacity of HCC cells.PSF1 maybe is a therapy target for the treatment of liver cancer in the future.
