The immune balance status of implanted gastric tumor model in rat on gene level are studied.Walker-256 cell lines are used to duplicate implanted tumor model in rat. The survival status of these rats was observed. Two weeks later, the mononuclear cell was separated from heart blood of these rats and was cultivated. RTPCR was used to test the quantity of the expression of GATA-3, T-bet, IL-4 and IFN-γ. The expression of GATA3 on implanted gastric tumor rat are higher than that on normal rat, and the expression of T-bet on implanted gastric tumor rat are lower than it on normal rat (P < 0.05 ). IL-4 were lower in tumor rat than that in normal rat, while IFN-Υwere higher than that in normal rat (P <0.05). There was a Th1/Th2 imbalance on gene level of Th cellspecific transcription factor and cytokine on implanted gastric tumor model in rat, and it was mainly Th2 subsets hyperactivity. The body of tumor rat was on an anti-tumor immunosuppressant status.%从基因水平了解种植性胃肿瘤鼠辅助性T细胞(Th)的免疫平衡状态.建立Walker-256种植性胃肿瘤大鼠模型,观察其生存状态,从心脏血中分离单个核细胞(MNC)进行细胞培养,用RT-PCR法检测GATA-3、T-bet、IL-4、IFN-γ基因表达水平.种植性胃肿瘤鼠较正常对照组GATA-3表达增高, T-bet表达降低(P<0.05),IL-4水平高于正常对照组,而IFN-γ水平低于正常对照组(P<0.05).种植性肿瘤大鼠存在Th细胞特异性转录因子、细胞因子基因水平的Th l / Th2平衡紊乱,以Th2亚群亢进为主,肿瘤鼠机体处于抗肿瘤免疫抑制状态.
