目的 观察HOX转录反义RNA(HOTAIR)在非小细胞肺癌(NSCLC)组织中的表达变化,并探讨其临床意义.方法 收集125例NSCLC患者的肿瘤组织及其瘤旁正常肺组织,采用实时RT-PCR法检测HOTAIR的表达.检索高通量基因表达数据库(GEO)、Array Express数据库中NSCLC组织与HOTAIR表达相关的芯片数据.结果 125例患者中,HOTAIR在NSCLC组织中的表达高于癌旁正常肺组织(P<0.05).HOTAIR表达水平与NSCLC患者年龄、肿瘤直径、淋巴结转移、TNM分期有关(P均<0.05).从数据库检索出的基因芯片显示HOTAIR表达水平与肿瘤病理类型不相关(P>0.05).结论 HOTAIR在NSCLC组织中呈现高表达,可能参与NSCLC的发生发展.%Objective To observe the expression changes of HOX transcript antisense RNA ( HOTAIR) in the non-small-cell lung cancer ( NSCLC) tissues and to investigate its clinical significance .Methods NSCLC tissues and adjacent tissues from 125 patients were collected and the expression of HOTAIR was detected by RT-PCR.Microarray data on the correlations between HOTAIR expression and NSCLC tissues were retrieved from Gene Expression Omnibus (GEO) and Ar-ray Express database .Results The expression of HOTAIR in the NSCLC was significantly higher than that in the adjacent tissues (P<0.05).The expression of HOTAIR was closely related to age , tumor diameter, lymph node metastasis and TNM stage of NSCLC patients (all P<0.05).The microarray data from GEO Profiles demonstrated that the HOTAIR ex-pression level was not related with pathological pattern (P>0.05).Conclusion HOTAIR is highly expressed in the NSCLC tissues , and its expression may take part in the occurrence and development of NSCLC .
