雷公藤内酯醇对耐顺铂人卵巢癌细胞株COC1/DDP增殖和凋亡的影响

         

摘要

目的 观察雷公藤内酯醇(TP)对耐顺铂(DDP)人卵巢癌细胞株COC1/DDP细胞增殖和凋亡的影响.方法 用10 ng/ml TP(TP组)培养COC1/DDP细胞24、48、72 h,每组设3个复孔及3个对照孔.采MTT法检测两组细胞的细胞增殖抑制率.用透射电镜观察培养24 hTP组细胞的凋亡形态,用DNA电泳分析细胞DNA凋亡状况.结果 TP组COC1/DDP细胞增殖抑制率且呈时间依赖性提高(P均<0.05).作用24 h后,透射电镜观察可见TP组COC1/DDP细胞表面微绒毛消失,细胞核固缩、染色质凝集成新月状,有凋亡小体形成.DNA电泳可见TP组DNA断裂形成"梯形"条带.结论 10 ng/ml TP对COC1/DDP细胞有明显的抑制增殖和促凋亡作用.%Objective To study the effects of triptolide (TP) on prolifration and apoptosis of cisplatin-resistance human ovarian cancer line COC1/DDP. Methods MTT assay was used to analyse growth inhibition effect of DDP(0.5μg/ml) and medium concentration TP ( 10 ng/ml) on COC1/DDP cells. 24 hours after the treatment of TP, the COC1/DDP ultrastructure by transmission electron microscopesh was detected. The apoptosis fracture zone investigated with DNA electrophoresis. Results Medium concertration TP could evidently inhibit the growth of COC1/DDP cells,and inhibitory effect on COC1/DDP in a time-dependent manner(P <0.05). 24 hours after the treatment of TP with DDP, microvillus on the surface of COC1/DDP cells disappeared, cell-nucleus was condensed, and chromosomal was agglutinated into crescent, apoptosis body had formed. DNA trapezoid strips could be seen with agarose gelelectrophoresis. Conclusions 10 ng/ml TP can inhibit the growth and promote the apoptosis of COC1/DDP.
