Objective To investigate the effect and mechanism of β-elemene on apoptosis in human gastric cancer MGC803 cells. Methods Human gastric cancer MGC803 cells were treated with different concentrations of β-elemene for 24 ~48 h, and the cell viability was measured by MIT assay, apoptosis was evaluated by flowcytometry following propidium iodide (PI) staining. Western blot was employed to detect the expression of phosphor-extracellular signal-regulated kinases (ERK) and total ERK after the cells were treated with β-elemene. Results β-elemene inhibited the viability of MGC803 cells in a dose-dependent manner. Further study showed that β-elemene induced apoptosis and inhibited ERK activity.Conclusion β-elemene can induce apoptosis in human gastric cancer MGC803 cells probably by inhibiting ERK signaling pathway.%目的 观察β-榄香烯对胃癌MGC803细胞凋亡的影响,并探讨其机制.方法 用不同浓度β-榄香烯作用于MGC803 24~48 h,MTT法检测细胞活力,流式细胞术测算细胞凋亡率,Western blot法检测MGC803中的磷酸化细胞外信号调节激酶(p-ERK)及总ERK.结果 随着药物浓度增加,细胞存活率逐渐降低(P均<0.05),细胞凋亡率明显增高(P均<0.05);随着药物浓度增加、作用时间延长,MGC803中p-ERK表达量显著下降(P均<0.05).结论 β-榄香烯可诱导胃癌MGC803凋亡,其机制可能与抑制细胞ERK通路活性有关.
