Objective To investigate the influence of bone morphogenetic protein microspheres on the proliferation and the expression of marrow stromal cell of canine. Methods Bone morphogenetic protein was enwrapped in the microspheres,which was prepared with PLA and PGA. The influences of bone morphogenetic protein microspheres on the proliferation, expression of type Ⅰ collagen of marrow stromal cell of canine were investigated by histochemistry and cell culture. Result Bone morphogenetic protein microspheres could significantly stimulate alkaline phosphatase activity and I collagen staining increased, but had no effect on the marrow stromal cell proliferation in vitro culture. Conclusion Bone morphogenetic protein can increase the osteogenic capacity of marrow stromal cell in vitro with the micropheres made of PLA and PGA as the carrier.%目的 观察骨形态发生蛋白微球对犬骨髓基质细胞(MSC)增殖及分化的影响.方法 聚乳酸聚乙醇酸共聚物制备微球,采用细胞培养及组织化学等方法观察骨形态发生蛋白载药微球对犬MSC增殖、分化及Ⅰ型胶原表达的影响.结果 骨形态发生蛋白微球对MSC的增殖无明显影响,但对细胞的分化功能及Ⅰ型胶原表达有明显的促进作用.结论 骨形态发生蛋白能够提高MSC的体外成骨能力,且微球可作为骨形态发生蛋白的良好载体.
