hTERT siRNA对宫颈癌Hela细胞增殖及化疗敏感性的影响

         

摘要

Objective To investigate the effects of hTERT siRNA targeting human telomerase reverse transcriptase (hTERT) gene on the proliferative activity of Hela cells and chemotherapy sensitivity. Methods Experiment was divided into hTERT siRNA group, Control siRNA group, empty vector group and the blank cell group, hTERT siRNA, Control siRNA and empty vector were respectively transfected into cervical cancer Hela cells. Expression of hTERT mRNA and pro-tein in Hela cells were detected by Real time-PCR and Western blot. Cell proliferation at 24, 48, 72, 96 h were detected by MTT assay. Treating stable transfection cells of these groups with different concentrations of cisplatin and bleomycin, cell viability and IC50, were measured by MTT assay. Results Compared with the other groups, hTERT mRNA and protein expression level were much lower (P < 0.05 ) , cell growth was inhibited and IC50 concentration of cisplatin and bleomycin significantly decreased in the hTERT siRNA group. Conclusion hTERT siRNA can significantly inhibit the expression of hTERT gene and the cell proliferation in cervical cancer Hela cells, and also increase the sensitivity of cells to chemothera-peutic drugs.%目的 探讨靶向人端粒酶反转录酶(hTERT)siRNA对宫颈癌Hela细胞增殖及化疗敏感性的影响.方法实验分为hTERT siRNA组、Control siRNA组、空载体组及空白细胞组,分别以hTERT siRNA、Control siRNA及空载体转染宫颈癌Hela细胞.采用real time-PCR法及Western blot法检测各组细胞hTERT mRNA及蛋白表达,MTT法检测24、48、72、96h细胞增殖活性.并对上述各组细胞分别用不同浓度顺铂、博来霉素处理,MTT法检测细胞存活率及两种化疗药物的IC50.结果 与其余各组相比,hTERT siRNA组hTERT mRNA及蛋白水平表达明显降低(P<0.05),细胞增殖受到抑制,顺铂、博来霉素的IC50显著下降.结论 hTERT siRNA可显著抑制宫颈癌Hela细胞中hTERT基因的表达,抑制细胞增殖,并提高细胞对化疗药物的敏感性.
