Objective To investigate the association of TGF-β1 gene single-nucleotide polymorphisms with susceptibility and recurrence of nasopharyngeal carcinoma. Methods One hundred and ninty-three historically di-agnosed and followed nasopharyngeal carcinoma patients and 231 healthy people were included. After collecting the peripheral blood samples for all, single nucleotide polymorphisms were analyzed by Taqman real-time PCR. Genotypes of all samples were analyzed with SDS program. All statistical analyses were performed with statistical product and service solutions v13.0. OR value and 95% confidence interval ( CI ) were calculated. Results Single factor analysis showed that TGF-β1-869 genotype has no relationship with the susceptibility of nasopharyngeal car-cinoma; the same as the result of TGF-β1-509 genotype, there was no relationship with susceptibility of nasopha-ryngeal carcinoma either. Multiple factors analysis considering age, gender and smoking also showed that both TGF-β1-869 and TGF-β1-509 genotype didn't increase the risk of nasopharyngeal carcinoma significantly. After further univariate and multivariate analysis, we didn't find any association between every genotype of TGF-β1-869 and TGF-β1-509 and recurrence of nasopharyngeal carcinoma, P value has no significant difference. Conclusion TGF-β1 genes polymorphism have no relation with the susceptibility and recurrence of nasopharyngeal carcinoma.%目的 探讨转化生长因子(TGF)-β1-869T/C,TGF-β1-509C/T基因单核苷酸多态性(SNPs)与鼻咽癌易感性与复发的关系.方法 采用病例对照研究,采集来自辽宁地区不同医院的鼻咽癌患者193例,同期就诊的非肿瘤患者231例静脉血,酚氯仿法提取基因,Taqman real-time聚合酶链反应(PCR)方法及SDS软件对TGF-β1基因型进行分型.SPSS 13.0软件包进行数据分析,以χ2检验比较SNPs基因型在病例与对照之间分布的差异,以单因素和多因素Logistic回归计算比值比(OR)及95%可信区间(CI).结果 单因素分析显示,TGF-β1-869位点的各基因型与鼻咽癌易感性不相关,同样TGF-β1-509位点的各基因型与鼻咽癌易感性也不相关,经校正年龄性别吸烟状态后显示仍与鼻咽癌易感性仍不相关.经单因素及多因素进一步分析后显示:TGF-β1-869、TGF-β1-509位点各基因型与鼻咽癌复发也不相关,差异无统计学意义.结论 TGF-β1-869和TGF-β1-509位点SNPs与鼻咽癌易感性及复发均不相关.
