目的:探讨提高孕妇外周血中游离胎儿 DNA产量的新方法。方法用QIAamp DNA Blood Mini Kit和QIAamp Circulating Nucleic Acid Kit提取56名孕妇外周血血浆DNA。应用荧光定量聚合酶链反应(PCR)分析样本中游离胎儿DNA的含量。结果与QIAamp DNA Blood Mini Kit相比,经QIAamp Circulating Nucleic Acid Kit提取试剂盒提取的总DNA平均浓度是前者的1.5倍。29名早期妊娠孕妇外周血游离胎儿DNA平均浓度为812拷贝/mL ,27例晚期妊娠为4876拷贝/mL。29名男性胎儿SRY基因平均浓度是前者1.2倍,妊娠早期平均浓度为28.6拷贝/mL ,晚期平均浓度为264拷贝/mL。结论经改良后,采用QIAamp Circulating Nucleic Acid Kit提取试剂盒可以获得更高浓度的胎儿游离 DNA ,可以应用于孕早期和孕中期的无创性产前基因诊断。%Objective To investigate a new method that can improve the yield of fetal cell free DNA .Meth-ods Fetal cell free DNA was extracted from 56 maternal plasma by QIAamp DNA Blood Mini Kit and QIAamp Circulating Nucleic Acid Kit .Results Compared with QIAamp DNA Blood Mini Kit ,the average concentration of total DNA extracted by QIAamp Circulating Nucleic Acid Kit were 1.5 times .Twenty-nine cases of early preg-nancy were 812 copies/mL ,27 cases of late pregnancy is 4 876 copies/mL .Average concentration of 29 cases of male fetal SRY gene were 1.2 times of QIAamp DNA Blood Mini Kit .Average concentration in early pregnancy is 28.6 copies/mL ,that of late is 264 copies/mL .Conclusion We can get a higher concentration of fetal cell free DNA using modified QIAamp Circulating Nucleic Acid Kit .It can be applied in non-invasive prenatal diagnosis of early and mid-term pregnancy .
