为获得适宜的再生体系和光皮树单倍体植株,以光皮树优良无性系花药为外植体,采用正交设计方法,研究不同消毒方法对外植体的消毒效果、不同培养基类型对愈伤组织诱导的影响.结果表明:0.1%升汞消毒5 min效果最为理想;MS +1.5 mg/ L 2,4-D+1.0 mg/L NAA+O.8 mg/L BA+30 g/l.蔗糖、MS +1.0 mg/L 2,4-D+O.5 mg/L NAA+O.4 mg/L BA+30 g/L蔗糖培养基均能获得较高的愈伤组织诱导率和愈伤组织增殖率;前者愈伤组织结构疏松,呈白色;而后者愈伤组织结构紧实,呈浅黄色.%In order to obtain optimal regeneration system and some haploid plants, taking anthers form superior clones in Swida wilsoniana as explants, effects of different disinfection methods and medium types on anther callus induction were researched by the mean of orthogonal design. The results showed that it was an ideal disinfection method to soak explants in 0. 1 % HgCl2 for 5 minutes. Higher callus induction ratio and proliferation ratio could be obtained in MS supplemented with 1.5 mg/L 2,4-D, 1.0 m.g/L NAA, 0. 8 mg/L BA and 30 g/L sucrose or with 1.0 mg/L 2,4-D, 0. 5 mg/L NAA, 0. 4 mg/L BA and 30 g/L sucrose. However, the callus induced from the former medium was white with loose structure, and the callus induced from the latter medium was flavescent with compactness structure.
