IGF -1在不同鸡与鹌鹑杂交禽胚胎肌肉发育过程中的表达规律与差异

         

摘要

[目的]研究IGF -1基因在蛋用型种鸡(♂)×鹌鹑(♀)和肉用型种鸡(♂)×鹌鹑(♀)两种不同杂交禽胚胎肌肉发育过程中的表达规律,比较该基因在两种不同杂交禽中的差异表达,探讨IGF -1基因在肌肉发育网络调控中的作用.[方法]通过人工受精的方法,蛋用型种鸡(♂)×鹌鹑(♀)获得蛋杂交禽蛋、肉用型种鸡(♂)×鹌鹑(♀)获得肉杂交禽蛋,与鸡蛋、鹌鹑蛋按照鸡标准孵化条件同批入孵,采集第7~17 d活胚胸肌组织,应用实时荧光定量PCR技术检测IGF -1基因在四个物种中每个时间点mRNA的相对表达量.[结果]IGF -1基因在胚胎肌肉发育的第7~17 d均有表达,在第10 d达到第一次峰值,鸡和鹌鹑在第16 d达到第二次峰值,蛋杂交禽和肉杂交禽在第15 d达到峰值.[结论]IGF -1可能在胚胎肌肉的生长发育过程中起调控作用;IGF -1 mRNA表达规律与成肌细胞增殖和肌管融合规律一致,可能参与调控成肌细胞的增殖和肌管的融合.%[Objective]The purpose of this study was to analyze IGF - 1 mRNA expression profiles and differences during embryonic myogenesis in two strains of hybrid of chicken - quail, and evaluate the potential relationships between IGF - 1 expression and myogenesis. [ Method] Two strains of cross - bred eggs of quail ( ♀ ) were acquired by artificial insemination by chicken( ♂ ) -semen, with the eggs of chicken, quail and hybrid in one batch hatched according to the standard condition of chicken, and then the breast muscle tissues from day 7 to day 17 were collected. The expression of IGF - 1 mRNA in different species every day was revealed by real - time quantitative PCR. [Result]The expression of IGF - 1 mRNA was detected from day 7 to day 17. The first peak time of IGF - 1 expression was noticed at day 10 in four species, the second peak time was noticed at day 16 in Chicken and Quail, and at day 15 in Meat - Hybrid and Egg - Hybrid. [ Conclusion] IGF - 1 maybe plays a key role in the process of myogenesis. The expression profile of IGF -1 was in accord with the profile of myoblasts proliferation and muscle tube fusion, and it maybe plays a key role in the process of myoblasts proliferation and muscle tube fusion.
