瘦素及瘦素受体与子宫内膜异位症关系分析

         

摘要

目的 探讨瘦素与瘦素受体在子宫内膜异位囊肿中的水平以及其在正常育龄妇女卵巢组织中与子宫内膜异位症患者卵巢组织中表达的差异.方法 选取2013年7月至2015年3月32例子宫内膜异位症患者作为内异组,并选取同期行子宫切除术的32例妇女作为对照组,分别采集两组人员的血样、腹腔液(PF)以及卵巢组织,内异组患者在进行外科手术时取用子宫内膜异位囊肿组织(OE)并吸取囊肿液(EF)以及腹膜活检组织(PI),然后采用ELISA试剂盒检测瘦素水平以及免疫印迹法测定瘦素受体的表达情况.结果 瘦素受体在正常卵巢组织中与子宫内膜异位症患者卵巢组织中的表达差异明显(P<0.05),但是瘦素水平之间差异不显著(P>0.05).瘦素和瘦素受体在子宫内膜异位症患者PI中的表达均无明显差异(P>0.05).酶联免疫吸附剂测定结果显示,子宫内膜异位OE中的瘦素水平明显高于血清以及PF中的水平.内异组患者子宫内膜异位OE和PI中瘦素与瘦素受体表达水平具有较强的相关性.PF中瘦素水平与PI中瘦素以及瘦素受表达之间具有强烈的相关性.子宫内膜异位EF中瘦素水平与子宫内膜异位OE中瘦素以及瘦素受体表达之间具有强烈的相关性.结论 瘦素通过与瘦素受体之间的相互调节作用在子宫内膜异位囊肿的发病过程中起重要的作用.%Objective To investigate the differences between leptin and leptin receptor in endometriosis in women of childbearing age and their expression in normal ovarian tissue or endometriosis patients with infertility ovarian tissue. Methods From July 2013 to March 2015, 32 cases of endometriosis patients were selected as the observation group,and the same period 32 cases of tubal ligation for women were chosen as a control group in our hospital, they were taken blood,peritoneal fluid,and ovarian tissue. The observation group were drawn during surgery cyst and cyst fluid lessons and peritoneum biopsy and then the expression of leptin receptor were measured using ELISA kit leptin levels and immunoblotting. Results The expression of leptin receptor in ovarian tissue of normal ovarian tissue was significantly different from that in endometriosis(P<0.05),but there was no significant difference between leptin levels(P>0.05). There were no significant differences in the expression of leptin and leptin receptor in PI in patients with endometriosis(P>0.05). The results of enzyme-linked immunosorbent assay showed that the levels of leptin in endometriosis OE were significantly higher than those in serum and PF. The expression levels of leptin and leptin receptor in endometriosis OE and PI were significantly correlated in endometriosis group. There was a strong correlation between leptin levels in PF and the expression of leptin and leptin in PI. There was a strong correlation between the levels of leptin in the endometriosis EF and the expression of leptin and leptin receptor in endometriosis OE. Conclusion Leptin action through mutual adjustment between leptin receptor plays a very important role in the pathogenesis of endometriosis.
